We propose that the multiple-choice decision-making process can be approximately divided into two phases: (1) the coarse selection; and (2) fine selection. Notably, opposite-feature inhibition operates on a much wider range than similar-feature inhibition in our model. After motion onset, the activities of neural pools located around the direction of each target build up over time, and the opposite-feature inhibition predominates the network dynamics. The information encoded in the stimulus input is mainly used to screen out unfavorable options one by one, until few adjacent alternatives are to be differentiated. Then, the similar-feature inhibition begins to have a role. This narrow inhibition operates locally to ensure that the bump of activity profile appears only around the direction of one target. The relative strength of opposite-feature versus similar-feature inhibition affects the transition between the two phases.

The sample sizes and statistical tests were selected based on previous studies with similar methodologies. All experiments were repeated at least three times, giving similar results. The results of independent experiments are presented as mean values; error bars represent the SD or the propagated error when the value of each experiment was itself calculated as a mean of individual cells. Statistical significance was tested using the two-tailed t test.

Prometaphase-arrested cells were harvested by shake-off and mitotic cell extracts were treated with or without phosphatase inhibitor for 30 min at 30°C. Histone isolation was followed by Western blot analysis of pT120-H2A. Equal loading was monitored by Ponceau S staining.
For EPSCs, a detailed state model of AMPA receptors postsynaptic to AN terminals on bushy neurons of the nucleus magnocellularis of the chicken (Raman and Trussell, 1992) was adapted for this study. These neurons are homologous to mammalian bushy cells, and they have very similar synaptic conductances (Gardner et al, 2001). The time course of the receptor model was adjusted to match that of mouse bushy cells, by allowing the opening and closing rates, Ro1, Ro2, Rc1, and Rc2, of the model to vary [see Table 2; all other parameters remained as described (Raman and Trussell, 1992)], with an assumed Q10 of 1/5 (Spirou et al, 2005).

Prior to, or following the fear conditioning and extinction assay, mice underwent real-time place preference assay, counter-balanced between groups. Mice were connected to optical fibers and individually placed in a rectangular enclosure with two chambers (28 cm x 28 cm each), and allowed to freely explore the two chambers for 20 min. Gray-scaled video was recorded at 30 fps and mouse location and velocity was tracked throughout experiment using Ethovision XT 9. For the first 5 min, the mice freely explored both chambers but they did not receive optical stimulation (‘baseline’). For the next 15 min, mice continued to have access to both chambers, but the presence in one chamber resulted in continuous delivery of laser light (594 nm, 6 mW, Cobalt) to the implanted optic fibers. The ‘light on’ chamber was randomly assigned and counterbalanced within each experimental group (NpHR or YFP).
FRET analyses were carried out by excitation with a 440 nm diode laser and by recording of CFP (CFP signal) and YFP (FRET signal) fluorescence emission in z-stacks. Background-corrected FRET ratios (CFP signal/FRET signal) were calculated in ImageJ using the Ratio Plus plugin. FRAP analysis of EGFP-Bub1 was performed with a 488 diode laser on one KT pair per cell. Overall bleaching was corrected using the signal intensities at a cytoplasmic region not targeted for photobleaching (average of the first 4 frames). Fluorescence recovery half-times and plateaus were determined by non-linear curve fitting based on a one-phase association in Prism software (GraphPad).
The increasing findings and improved understanding of the microbicidal properties of CDots have paved the way toward the design of a multifunctional CDots-based theranostic platform for simultaneously integrating the diagnostic functions and therapeutic effects. A few studies have already been reported on using CDots for antimicrobial theranostics to achieve the concurrent effects of bioimaging, drug delivery and release, and photodynamic therapy [90, 96-98]. For example, Mitra, et al. coated CDots onto the surface of ZnO nanorod for antibacterial and bioimaging applications . The resulting nanohybrid could be taken up by S. aureus cell to display bright green fluorescence emissions, also exhibiting antibacterial activities against S. aureus and E. coli in a concentration dependent fashion . Thakur, et al. reported that CDots could be used simultaneously as a tracer to exploit the photoluminescence properties and as a vehicle to carry and deliver a significant amount of therapeutic agents such as ciprofloxacin for potential theranostics . Dou, et al. explored CDots for both antibacterial activities and gene delivery capabilities, and observed much enhanced gene transfection efficiency over that of the naked DNA delivery . Pramanik, et al. febricated multi-component nanostructures by combining fluorescent CDots, magnetic nanoparticles, and pardaxin antimicrobial peptides, which were designed for the diagnosis, selective separation, and early disinfection of pathogens on the same material platform . In another investigation , CDots-like nanostructures were used as two-photon contrast probes for the tracking and localization purposes in the two-photon imaging in a three-dimensional biological environment and concurrently for the generation of ROS to eliminate MDR bacterial species.

Radiation is a current standard treatment of glioma. The fractionated radiotherapy with low dose of radiation over weeks has been employed in glioma patients, while radiotherapy can only offer palliation due to the radioresistance. We cumulatively radiated a glioblastoma cell line, U87MG, and screened radioresistant glioma cells. A transcriptome sequencing was performed to analyze the transcription differences between the raidoresistant and control cells, which showed the mitochondria NADH–ubiquinone oxidoreductase (Complex I) subunits were up-regulated in the radioresistant cells. The copy numbers of mitochondria were increased in the radioresistant glioma cells. After using mitochondria Complex I inhibitors, rotenone and metformin, to treat glioma cells, we found the resistant glioma cells re-sensitized to radiation. These results demonstrate that Complex I is associated with the fractioned radiation-induced radioresistance of glioma and would be a potent target for clinical radiotherapy of glioma.
Rapamycin (LC Laboratories) was dissolved in DMSO at 1 mg/ml and used at a final concentration of 200 nM. PalmC was dissolved in DMSO at 10 mM and used at 5 µM. The Flipper-TR probe, synthetized following reported procedures (Colom et al, 2021), was dissolved in DMSO and used at a final concentration of 2n g/ml. Latrunculin A (L5163; Sigma-Aldrich) and Phalloidin (A1488; Applichem) were dissolved in ethanol at 2 mM and 1 mg/ml respectively, and used at 1 µM.
Inhibition of Bub1 affects chromatid cohesion
This is an important feature, indicating that the neural activity not only encodes the information about the choice itself but also reflects the likelihood of a decision to be correct, as suggested by theoretical studies (Ma et al, 2006; Beck et al, 2008). This also implies that the more similarity two neural pools share in their preferred directions, the more correlated their activities become. This similarity effect mainly stems from opposite-feature inhibition, which leads to stronger lateral inhibition between pyramidal cells with larger angular distance. Consequently, two excitatory neural pools with opposite preferred directions tend to repress each other remarkably, which intensifies the competition between them. Thus, their firing rates, like R0 and R180, are most anticorrelated during decision-making. As the winning neural pool builds up its activity, the neural pool with opposite preferred directions has the lowest buildup rate.

If you create an FSP Resource and do not control it with an AF resource, you are merely backing up to a local directory. Although a fine method for doing quick backups, BackupEDGE cannot use the local directories for disaster recovery.
Cells were seeded into 96-well plates at aconcentration of 5–6×104 cells/ml. Cell viability wasevaluated with a3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT)assay. The MTT assay was performed as follows: 20 μl of 5 mg/ml MTTdissolved in phosphate-buffered saline (PBS) was added to eachindividual well followed by incubation at 37°C for 4 h. Thesolution was then removed, and the produced formazan wassolubilized in 100 μl dimethyl sulfoxide (DMSO). Absorbance wasmeasured at 570 nm using an automated microplate reader (Bio-RadLaboratories, Hercules, CA, USA). Cell viability was expressed as apercentage of the control culture value.
The median and quartiles for each mutation group are indicated. P values refer to intergroup statistical analysis of log-transformed data by the Student t test, with significant differences in lestaurtinib IC50 distribution being seen between WT and ITD cases and between WT cases and all FLT3 mutants grouped together.
We found that inhibition of the lateral VTA at the tone offset did not influence extinction learning of the preceding tone. This was consistent with the previous observation that inhibition of a subpopulation of VTA neurons with projections to NAc Core did not affect extinction (Luo et al, 2021). However, similar to medial VTA, this manipulation increased freezing in the timepoints directly after the inhibition, suggesting that despite the fact that these neurons did not regulate learning about the value of a preceding event (the tone), they did affect behavior in the time period subsequent to the manipulation. Consistent with this manipulation affecting subsequent timepoints, inhibition at the tone onset, when these neurons have a burst of activity, slowed the rate of extinction. However, we cannot rule out that this manipulation affected the expression of freezing rather than extinction learning. Additionally, this lateral VTA tone onset inhibition had more subtle effects on behavior relative tomedial VTA tone offset inhibition.
Two-Photon Photoexcited Photodynamic Therapy and Contrast Agent with Antimicrobial Graphene Quantum Dots

In a first series of experiments, we used time-lapse imaging to compare progression through mitosis in asynchronously growing HeLa and RPE1 cells in response to either Bub1 inhibition or siRNA-mediated Bub1 depletion. In line with previous results (Kitajima et al, 2005; Tang et al, 2004b), depletion of Bub1 from HeLa cells significantly prolonged duration of mitosis, due to delayed chromosome alignment and delays in prometa- and metaphase (Figure 2A and C, Figure 2—figure supplement 1A).
Dual AT-MT resulted in highest tumor growth inhibition
Equal amounts of protein from eachsample (40 μg) were resolved by sodium dodecyl sulfate(SDS)-polyacrylamide gel electrophoresis (12%) and transferred ontoPVDF membranes. The membranes were incubated in blocking buffer(non-fat milk) and then incubated overnight at 4°C with rabbitpolyclonal antibodies against Bax (CST, 1:1,000, 20 kDa), Bcl-2(CST, 1:1,000, 26 kDa) or β-actin (1:4,000, 43 kDa) (Beyotime). Themembranes were stringently washed and incubated with HRP-conjugatedsecondary antibodies (Proteintech Group, Chicago, IL, USA), for 1 hat room temperature. After washing, proteins were detected usingenhanced chemiluminescence, and images were acquired using aBio-Image Analysis System (Bio-Rad Laboratories).

OGD/R was performed with the AnaeroPack system,which includes a rectangular container (9/5×6/75×3/25 in; 2/5 l)and one AnaeroPack sachet. Briefly, SH-SY5Y cells were plated inDMEM. After treatment, the cells were incubated in the sachet witha BBL disposable anaerobic indicator strip and placed into thecontainer. After 60 min of incubation, the oxygen concentration wasless than 1% and the CO2 concentration was approximately18%. After 12 h, the cells were incubated for 2 h for reoxygenationand glucose restoration. In the normoxia control group, the cellswere cultured with DMEM containing glucose under normalconditions.
This inhibition eliminates poorly timed spikes, resulting in improved spike timing on a millisecond timescale
The Mount Device Node is the device node that you will use to mount this attached filesystem, such as `/dev/sdb1'. The Exclude Node is the device node that will be excluded by RecoverEDGE during disk preparation for disaster recovery. The idea is that you do not want RecoverEDGE re-creating the filesystem on your removable hard drive that previously held the backup you wanted to restore from. The Exclude Node should be the `whole disk' node, even if you are mounting only a partition of it. For example, if the Mount Device Node is `/dev/sdb1', you would want to use `/dev/sdb' as the Exclude Node.

The nanoparticles were further labeled with IRDye800CW and Gd-DTPA to facilitate MRI/NIRF imaging for non-invasive detection of PD-L1 activity. In vivo, T1-weighted MRI and NIRF imaging demonstrated significantly enhanced accumulation of PD-L1 antibody-conjugated nanoparticles in mice bearing 4T1 and CT26 tumors. In addition, the targeted nanoparticles demonstrated significantly enhanced antitumor activity compared to nontargeted nanoparticles, free PTX, and free PD-L1 antibody. The results suggested that immunotherapy functioned as an adjuvant to improve the efficacy of chemotherapy. In another study, a nanoplatform consisting of SPIONs, anti-PD-1 antibody, perfluoropentane, and GRGDS peptides showed effective ablation of melanoma with combination immunotherapy and PTT . Finally, Liu et al. designed iron oxide nanorings for combined immunotherapy and MHT to eradicate primary tumors and inhibit metastatic spread . Mild MHT activated the host immune system and inhibited the immunosuppressive tumor response through PD-L1 blockage. With the recent approval of anti-PD-L1 antibodies for the treatment of melanoma, methods to quantify PD-L1 expression for treatment planning and to monitor treatment success will be important .
Depletion of Bub1 is known to weaken SAC signaling in human cells (Klebig et al, 2009; Meraldi and Sorger, 2005; Perera et al, 2007). To test the impact of Bub1 catalytic activity on SAC function, we first analyzed KT levels of Mad1, Mad2 and BubR1 in BAY-320 or BAY-524 treated cells. With the possible exception of a very minor effect on BubR1, the localization of none of these SAC proteins was significantly affected by Bub1 inhibition (Figure 6A and B, Figure 6—figure supplement 1A). In sharp contrast, and in agreement with previous reports (Boyarchuk et al, 2007; Johnson et al, 2004; Overlack et al, 2021; Sharp-Baker and Chen, 2001), Bub1 depletion decreased KT recruitment of all three proteins by 80–90% (Figure 6A and B, Figure 6—figure supplement 1B). Thus, the recruitment of several SAC components to KTs strongly depends on Bub1 protein, but not Bub1 kinase activity.

This works fine with disaster recovery. In other words, the attached storage device should be defined as a resource only on the machine to which the storage is physically attached. If that resource is an FSP with an AF, then it may be used for disaster recovery by that machine, and by remote machines. If it is an FSP without an AF, then it can be used for disaster recovery by remote machines only.
In contrast, the Basic Schedule editor does not let you select anything other than the Sequence onsite_system (for on-site full system backups). It also does not provide an option to perform Differential or Incremental Backups.
Visual stimuli were created using customized MATLAB software (courtesy of J. Bergan, Harvard University, Cambridge, MA) and presented (Mitsubishi XD300U projector) on a calibrated tangent screen at a distance of 35 cm from the eyes. The owl was positioned so that the visual axes were in the horizontal plane aligned with 0° elevation and 0° azimuth of the screen. All locations are given in double pole coordinates of azimuth, relative to the midsagittal plane, and elevation, relative to the visual plane.
BAY-320 was screenedin vitro, at 10 µM and 10 µM ATP, against a panel of 222 kinases using the Eurofins kinase profiler screen (Millipore). In addition, BAY-320 was screened, at 300 and 1000 nM, in an active site-directed competition-binding assay measuring 403 human kinases (Lead Hunter, DiscoverX Kinome Scan).

BackupEDGE is removed by typing custom from a character interface or running Software Manager from the GUI or scoadmin. From the full-screen interface, make sure that BackupEDGE for SCO OpenServer 6 is highlighted, then use the Software -> Remove Software option. Alternately, you may run custom from the command line. The following command will remove BackupEDGE for SCO OpenServer 6.
We introduce an open-source pipeline to automate freezing analysis of behavioral videos. The main motivation and advantage of this pipeline is automation of freezing analysis, even when mice are tethered to neural headgear - our analysis does not confuse headgear movement for mouse movement. Drawbacks to this technique include necessity for manual scoring to train the network (it took approximately seven hours to score the 33,000 images we used to train each network), and different networks must be trained for different backgrounds and neural headgear. Additionally, having a graphics processing unit (GPU) greatly speeds up network training (1–2 hrs to train 200 epochs on a 320 nVidia P100 GPU vs. 1 week on a 32 GB RAM, Intel Core i7 CPU) and using the network for analysis (10 min to run 25,000 frames on a 320 nVidia P100 GPU vs. 1 hr on a 32 GB RAM CPU).
Encryption algorithms use what are called "keys" to control the encryption and decryption of data. When one wants to encrypt data, one gives that data to the encryption algorithm, along with the "encryption key". The output of the algorithm is the encrypted data. Similarly, to decrypt the data, one provides the encrypted data along with the correct "decryption key" to the decryption algorithm to recover the original, unencrypted data. In some types of encryption algorithms, which are known as "symmetric ciphers", the encryption and decryption keys are identical. In other words, if one has the power (key) to encrypt data with a symmetric cipher, one also has the power (key) to decrypt it, and vice-versa. It is from this symmetry that this class of cipher gets its name. There is another class of ciphers, known as "asymmetric ciphers", in which two different keys are employed. Whatever is encrypted with one key cannot be decrypted by that same key. Rather, the other key must be used to decrypt it. This is a very important point to remember: in an asymmetric cipher, it is possible to encrypt a message without also having the ability to decrypt it, if only one key is known.
One of the most interesting effects of Bub1 depletion described so far relates to sister chromatid cohesion (Boyarchuk et al, 2007; Fernius and Hardwick, 2007; Kitajima et al, 2005; Tang et al, 2004b). In particular, depletion of Bub1 was shown to cause persistent arm cohesion and a redistribution of Sgo proteins from centromeres to chromosome arms (Kitajima et al, 2005). To directly demonstrate a role for Bub1 kinase activity in sister chromatid cohesion, we analyzed chromosome spreads prepared from mitotic HeLa cells or RPE1 cells after treatment with Bub1 inhibitors (Figure 3A, Figure 3—figure supplement 1A) or Bub1-specific siRNA for comparison (Figure 3B and Figure 3—figure supplement 1B).
Taken together, our pipeline provides an automatic, fast and effective method for scoring freezing. In this paper, we used the CNN to analyze over 500 hrs of behavioral data during fear conditioning and extinction, which would have been prohibitively time consuming without an automated approach.

Yet another difference between the classical and global inhibitory networks is in their connectivity. While all the neurons that mediate global inhibition must be mutually coupled in some way (either by mutual inhibition or by electrical synapses, see Results), all the neurons that mediate the classical inhibitory surround are not. Together, the preceding observations indicate the actions of two different inhibitory networks.
For those familiar with BackupEDGE 01/01/0x and earlier, this is behavior should be familiar. It is strongly recommended, however, that you do not use Expert backups, and instead use "Selective Restore" when using BackupEDGE 01/02/00 and later. If you are restoring files from a backup made with an pre-01/02/00 version of BackupEDGE, the filenames you type here should be the same as you would have typed in the older version of BackupEDGE. Optogenetic inhibition experiments further supports functional differences between dopamine neurons in the medial and lateral VTA. Posttranslational mechanisms may also contribute to receptor kinetics diversity within a cell population but seem unlikely to be the sole basis of the large difference in kinetics between bushy and T-stellate cells. It has been widely observed that, in other regions of the nervous system, glycinergic receptors composed of α2β subunits are abundant early in development and, with maturation, gradually switch to the adult α1β isoform. The trend is not always true in all neural circuits (Lynch, 2009). Our study suggests that such a switch might occur in T-stellate cells during development and might not occur in bushy cells, which would be consistent with the continued presence of α2 mRNA in the AVCN through adulthood (Piechotta et al, 2001).

Backups themselves are the most often used feature of any backup product. You may perform hundreds of backups before you ever have to restore a file, or perform a system recovery. Usually, most of the backups are Scheduled (unattended) backups. If you have not already done so, please review "Anatomy of a BackupEDGE Backup" on page 26 before reading further. BackupEDGE has two scheduling modes. The Basic Schedule mode very quickly creates a Scheduled Job for full system backups.
Expert Backup looks very similar to the Backup Multiple Files option, but in fact is very different. There are no reasons why this option would be used, except for troubleshooting purposes. When specifying files with Expert Backup, you are actually controlling how they will be named in the archive. If you use Relative Pathnames, the archive will use Relative Pathnames for those files. If you use Absolute Pathnames, so will the archive. If you mix and match Relative Pathnames and Absolute Pathnames, the archive will reflect this also. You will also be given the opportunity to select the Root Directory for the backup. Any relative file or Directory names to be included on the first line will be relative to that Directory. Filenames given on the second line should be specified as Absolute Pathnames, although the files listed in those files will be treated exactly as if you had typed them in on the top line manually. The Backup Multiple Files option is preferable to this method.

A series of modeling studies (Wang, 2002; Ma et al, 2006; Wong et al, 2007; Furman and Wang, 2008) was performed to explore the underlying mechanism for perceptual decision-making at the neuronal circuit level. Despite great progress made, some features of multiple-choice direction discrimination are not yet interpreted. One feature may be associated with elimination process, a common tactic adopted to rule out the least likely options to reduce the number of alternatives for inspection. Although several psychological models were built to explain this process (Tversky, 1972; Yaniv and Schul, 1997; Roe et al, 2001), a biophysically realistic model based on neural circuitry is still lacking.

In conclusion, GLGZD exerts protective effectsagainst glutamate-induced cellular injury. To our knowledge, thisis the first report revealing the role of GLGZD in protecting BV-2cells against glutamate-induced neurotoxicity. Further studies onmature primary neurons, as well as on animal models of Parkinson’sdisease and comparisons with known anti-parkinsonian agents are,however, required to establish both efficacy and safety. Based onthe protective effects of GLGZD on glutamate-induced cellularinjury, GLGZD may be used as a potential therapeutic candidate forthe treatment of neurodegenerative disorders.
HeLa S3 or RPE1 cells were incubated with kinase inhibitors or depleted of the indicated proteins for 48 hr. Cell suspensions were then fixed with 70% ice-cold ethanol and incubated with 0/2 mg/ml RNase (Sigma-Aldrich) and 5 μg/ml propidium iodide (Sigma-Aldrich). Cellular DNA content was determined by flow cytometry using FACSCanto II (BD Biosciences Clontech, San Jose, CA, USA) and FlowJo (Treestar, Ashland, OR, USA) instruments.
Record Locking The three possible options are Don't Lock Files, Unenforced Read, and Enforced Read, and deal with the way EDGEMENU handles files in use or locked by another program. Don't Lock Files makes no attempt to lock files, and waits if files are locked by another program. Unenforced Read places an advisory lock on each file while archiving it. It can archive files locked in this fashion by other programs, and files can be changed by applications while EDGEMENU uses this lock type. This is the default locking option. Enforced Read placed a hard lock on files to be archived. This is not usually recommended, and may cause deadlocks during a backup.
So far, only few substrates of Bub1 have been identified. Best characterized is the phosphorylation of histone H2A on threonine 120 (T120) (Kawashima et al, 2021). Phosphorylation of this site by Bub1 can be demonstrated not only in vitro but also in living cells (Kawashima et al, 2021; Lin et al, 2021; Sharp-Baker and Chen, 2001). Gua Lou Gui Zhi decoction (GLGZD), a traditional Chinese medicine consisting of different herbal medicines, has been used for centuries in the treatment of muscular spasticity following stroke, epilepsy or spinal cord injury. However, the precise mechanisms involved remain poorly understood. In the present study, we investigated the neuroprotective effects of GLGZD on glutamate-induced apoptosis in cultured BV-2 cells, as well as the underlying mechanisms. A 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was applied to assess the viability of the cells. An Annexin V/propidium iodide (PI) assay was utilized to analyze cellular apoptosis. Mitochondrial membrane potential (MMP) was evaluated by flow cytometry and laser scanning confocal microscopy. The gene and protein expression of the apoptosis-related genes, Bcl-2 and Bax, was analyzed by RT-PCR and western blot analysis, respectively. Furthermore, the expression of cleaved caspase-3 protein was detected by immunofluorescence. Glutamate treatment induced the loss of BV-2 cell viability, which was associated with an increase in the apoptotic rate, as well as an increase in the Bax/Bcl-2 ratio and the extracellular levels of cleaved caspase-3. Treatment with GLGZD significantly reversed these phenotypes, with its maximum protective effects observed at the concentration of 1,000 µg/ml.

Characterization of novel MPS1 inhibitors with preclinical anticancer activity
While it is true that many targeted receptors/biomarkers tend to be “overexpressed” on the surface of malignant cells compared to normal cells, they are certainly not unique and are often found in nonmalignant tissues as well. This is especially important in human patients where the ratio of the tumor mass to body mass is significantly lower compared to animal models; bringing into question the actual effectiveness of “active targeting” in clinical studies . A more effective measure might be to compare the expression of a receptor on a tissue rather than on a cellular level. In addition, it is likely that targeting ligands are only overexpressed on a small percentage of cancer cells, which results in heterogonous accumulation of nanoparticles in the tumor volume and sub-optimal targeting in some regions. Furthermore, while many of the targeting ligands result in cellular internalization, a more extensive evaluation of molecular targets should be based not only on expression but on the rate of internalization and the efficiency of endosomal escape . A more thorough understanding of the complex intracellular trafficking mechanisms to optimize the delivery of the therapeutic cargo to the desired destination is necessary to achieve the desired therapeutic effect. Another issue is that many promising receptors for active targeting are identified using cancer cell lines maintained in vitro, which may not be representative of what is present in a patient's tumor. A better strategy would be to correlate biomarker expression to tumor grade of human samples.
The adaptors Sla2 and Ent1/2 constitute a physical bridge between the PM and the actin cytoskeleton; each of these proteins possesses a PtdIns(4,5)P2-binding domain in its N-terminal part and, in the case of Sla2, a THATCH/talin-like domain in its C-terminal end (Skruzny et al, 2021). The Ent1–actin interaction domain is additionally known to be regulated by phosphorylation by kinases of the Ark family (Skruzny et al, 2021), which are also potential downstream targets of TORC2 (Rispal et al, 2021; Bourgoint et al, 2021).

Considering the role of Aurora B kinase in the regulation of KT-MT interactions and SAC signaling, the above results raised the question of what contributions Bub1 activity might possibly make to chromosome congression and/or the SAC. Although our initial analyses had not revealed a major impact of BAY-320 or BAY-524 on the overall timing of mitotic progression (Figure 2), we considered the possibility that inhibition of Bub1 might provoke compensatory effects on mitotic timing, notably a delay in congression and a concomitant acceleration of mitotic exit. According to such a scenario, effects on timing might conceivably cancel each other. In support of this possibility, we emphasize that the inhibition of mitotic kinases with pleiotropic functions have previously been shown to provoke opposing phenotypes (Santaguida et al, 2021; von Schubert et al, 2021). To explore the possibility of compensatory effects of Bub1 inhibition, we thus carried out more detailed analyses of mitotic progression, notably KT-recruitment of SAC components, SAC signaling and chromosome congression.

FSP backups cannot commence until the FSP Resource has been initialized one time. NOTE: Never place any other (non-BackupEDGE-created) files in the BackupEDGE directory on the FSP Resource. Owls were prepared for multiple electrophysiological experiments. Before the first experiment, an owl was anesthetized with isoflurane (1%) and a mixture of nitrous oxide and oxygen (45:55), and a head bolt was mounted at the rear of the skull. The Imc, on the other hand, contains large, GABAergic neurons that receive topographic input from the OT. Imc neurons project back broadly to the OTi-d in a manner consistent with the observed global inhibition. Moreover, many Imc neurons project within the Imc as well (Wang et al, 2004), consistent with our hypothesis of mutual inhibition among the neurons of the global inhibitory network. Another distinctive structural property of Imc neurons is that, although they project back widely to the OT, they specifically do not project back to the part of the OT from which they receive input (Wang et al, 2004). This “hole” in the back projection to the OT could account for the apparent lack of a divisive influence evoked by stimuli located inside the RF.

A Differential Backup1 follows the same Domain rules, but backs up only that data (files or Directories) that have been created or modified since the last successful Master Backup in the Sequence to which both backups belong. An Incremental Backup follows the same Domain rules, but backs up only those files or Directories that have been created or modified since the last successful Differential Backup or the last successful Incremental Backup, whichever is newer. A Master Backup has no dependencies on other backups; each one can restore all the data for a Domain to the point in time when that backup was made. A Differential Backup is dependent on the Master Backup it is based on. That is, it takes the Differential Backup plus the Master Backup on which it is based to restore all the data in a Domain to the state it was in when that Differential Backup was made. When a new Master Backup is made for a Sequence, any previous Differential Backups in that Sequence are no longer needed (unless you want to restore the Domain's data to a point prior to the last backup, of course). It will not impact the contents of any future Incremental Backup (see below) for the Sequence. You cannot perform a Differential Backup which will overwrite the Master Backup upon which it would be based. If BackupEDGE detects this, It will either Fail to do the backup, or Promote the backup to be a Master Backup, depending on the setting of the Promote A flag in the Advanced Settings window of the Scheduled Job.

There are many ways to use FSP and AF Resources. Some are compatible with RecoverEDGE disaster recovery, and some are not. It is important to understand which uses and devices server which purposes.
The responses to stationary dots rose to a peak value and then declined asymptotically to a steady-state value, following the classic difference-of-Gaussians response profile. Hence responses to stationary dots were fit just with the difference-of-Gaussians model.
Resource: A named set of properties describing one Device used by BackupEDGE. For example, a Resource may represent a tape drive, and include the appropriate Device Node(s) for it, the Hardware Block Size, default Edge Block Size, special commands needed for use with BackupEDGE, and so on. REV. The Iomega trade name for their line of storage devices and media using RRD, or Removable Rigid Disk, technology. Root: The Superuser, or System Administrator, of a UNIX or Linux system. The root user has complete system privileges. As most BackupEDGE tasks require this, full system backups and Scheduling require the user to be logged in as root. Scheduled Job: A Scheduled Job describes all of the actions necessary to archive a Domain through a single Sequence (in other words, perform a backup). Once defined, Jobs can be run via the Scheduler at prescribed times or directly from EDGEMENU. Sequence: An organizational unit for backups of a Domain. It is possible to create multiple Sequences which each backup the same Domain.
Use edgemenu:Admin->Define Resources to do this. Select `[NEW]', and use the right-arrow and left-arrow keys to change the resource type to `AWS'. This would typically done to create a separate Resource directory on S3 for use with a different schedule. Additional AWS entries may not be created until S3 Setup is complete.

Third, there is significant decrease in the baseline (before CS) on Extinction Day 1 and 2. It is unclear whether this affected the data analysis. Overall, although this is one of the most important results of this study, the data are not particularly strong.
You may FastSelect any current Scheduled Job, or create a new Scheduled Job, with or without using the Wizard. You may also Delete a Scheduled Job by pointing to it and pressing [F6] or [CTRL-X]. Each Scheduled Job is displayed with its name (simple_job), description (Basic Schedule), and its current status (Enabled to run at 23:00, although the list of which days is not given). We recommend selecting the [Backup], [Run Scheduled] option, choosing the Basic Schedule, and [Run]ning it. This takes one extra keystroke, but it runs the same nightly backup Scheduled Job that you defined during installation. It also keeps Sequence information intact.

Network mounts, such as NFS and CIFS/Samba, are excellent storage resources, but are NOT compatible with RecoverEDGE. They are useful for quick storage, targeted backups, and data migration, but RecoverEDGE boot media does not have the ability to mount and restore from them. For this reason FTP Backups are the preferred method for performing network backups. See "Configuring FTP Backups" on page 62 for more information.

If, for some reason, BackupEDGE cannot detect the volume size correctly, you may specify it directly in the Resource. However, if you do this, BackupEDGE will treat all media loaded into this drive as being the size given. Compression The only compression available is [S]oftware. This will cause the data stream to stop and start. Make sure you have a buffer under-run proof Device, or have set a large Record Buffer size or the Buffer Whole Disc flag if you use software compression.
When you press [Next] to save the resource, you will be asked if you want to `Initialize' it. You must let BackupEDGE initialize the resource. This tests the connection again and creates a control file named CTL in the destination directory. To initialize at a later time, use edgemenu:Admin->Initialize Medium. Note that initializing the resource will not erase any existing backups. If existing backups exits, the CTL file, which contains information about the individual archive segments, will be re-calculated.
Chromosomes are separated by an array of protein fibers called the mitotic spindle. A surveillance mechanism known as the spindle assembly checkpoint prevents the cell from dividing until all the chromosomes have properly attached to the spindle. A protein called Bub1 is a central element of the SAC. However, it was not clear whether Bub1 works primarily as an enzyme or as a scaffolding protein.

Inflammation is generally a key factor in the development of a variety of diseases, including atherosclerosis. The inflammasome is a multiprotein platform that activates procaspase-1 to caspase-1; in turn, caspase-1 can convert pro–interleukin (IL)-1β and pro–IL-18 into their bioactive secreted forms. Both IL-1β and IL-18 are major mediators of inflammation, and a number of laboratories have shown that both cytokines induce severe atherosclerotic lesions in mouse models. These findings suggest the involvement of the inflammasome in the development of atherosclerosis. NLRP3 is the most extensively studied inflammasome. It is activated in response to a variety of signals that are indicative of damage to the host, including tissue damage, metabolic stress, and infection. Cholesterol crystals are important inflammatory stimuli in atherosclerosis development acting as an endogenous danger signal via the NRLP3 inflammasome.

Verify / Index Archive All normal backup operations have default settings for performing verify passes after the backup, and also for creating index databases for FFR and IFR. You may use this option to verify and/or index the media at a later time. Prior to beginning, the media label will be read, and you may set verify level and change the relative Root Directory for bit-level verifies. By default, however, BackupEDGE will use the correct Root Directory to verify the archive. For users of older (pre-01/02/00) versions of BackupEDGE, this is different behavior. In those versions, you were responsible for selecting the right Root Directory. Verify (Only) Archive This shows the same fields and options as Verify / Index Archive, differing only in the choice of default setting (indexing is turned off by default). List Archive Contents This starts a listing without checking the media label first. There is no need to set the Working Directory for a listing. Show Archive Label This option will attempt to read and display the BackupEDGE label from the media.
The full dataset for each cell, representing responses to different frequencies and the recovery time courses, were all fit at one time (MATLAB function fmincon). Receptor desensitization was not included in this step, because it was modeled as part of the postsynaptic receptor mechanism. The fitting parameters were averaged from fits to individual cells and were applied uniformly to all synapses associated with a given parent terminal. The fit parameters are shown in Table 1 for each cell type and synaptic input.
Encryption provides the means of securing archive media against many types of intrusion, but it is not a "cure-all" for data security. Like any other tool it must be used correctly before it is effective. Even then, there are problems that it simply is not designed to solve. For example, one must consider the ways in which data could be received by unauthorized parties. If someone who has physical access to the machine(s) in question is determined to get the data, then he or she will almost certainly be successful. It does not matter in a case like this whether or not backups have sensitive data protected by encryption; the point of failure is not the backup media, but rather the original copy of the data! Similarly, if a user of a system is able to gain access as the root user, then there is no need to for him or her to attack the encrypted backups. Instead, he or she will simply copy the original data. Alternatively, important system programs could be replaced with malicious counterparts that record sensitive data over time, so that future intrusions are not even necessary! BackupEDGE itself might be replaced, or configured not to encrypt data.
Target-specific IPSC kinetics are essential for temporal processing
Figure 1A shows dose-response curves of the mean MTS responses to lestaurtinib and PKC412 treatment for all 65 evaluable samples, normalized to untreated controls and grouped according to FLT3 mutation status. At most concentrations studied, lestaurtinib was significantly more cytotoxic to primary AML blasts with FLT3 ITD mutations than to those that expressed only WT FLT3. There was also a trend toward greater drug sensitivity in blasts harboring FLT3 TKD-PMs. There was no evidence of a differential level of cytotoxic response to PKC412 according to FLT3 mutation status. For both drugs, cytotoxic responses of MNCs from different patients were highly heterogeneous, even after accounting for FLT3 mutation status. This variation in cytotoxic response was also independent of blast percentage in the MNC fraction, presenting WBC count, cytogenetic risk group, and French-American-British (FAB) classification. For each individual patient, Calcusyn software (Biosoft) was used to calculate a 50% inhibitory concentration (IC50) for cytotoxic responses to both lestaurtinib and PKC412. The heterogeneity of IC50 obtained, with marked overlap between FLT3 mutant and WT cases, is illustrated in Figure 1B.

Preparation of BAY-320 and BAY-524 inhibitors
To control for possible artifacts in these recordings, we recorded from DAT-Cre mice expressing Cre-dependent GFP (AAV5-DIO-eGFP or AAV5-DIO-eYFP) in medial or lateral VTA. There was little modulation in the signal in the control mice. The exception was the time of the shock, which generated depressed fluorescence, which was the opposite of the increased fluorescence observed with GCaMP6f in medial and lateral VTA (Figure 3—figure supplement 4). This suggests that our conclusions above were not due to a recording artifact.
Carbon dots (CDots) have emerged to represent a highly promising new platform for visible/natural light-activated microbicidal agents. In this article, the syntheses, structures, and properties of CDots are highlighted, representative studies on their activities against bacteria, fungi, and viruses reviewed, and the related mechanistic insights discussed. Also highlighted and discussed are the excellent opportunities for potentially extremely broad applications of this new platform, including theranostics uses.
BackupEDGE uses the concept of a `slot', similar to a tape library slot, to manage the archive segments. Every time you write a backup to a D2D device, you must select which slot to be used. Each archive is a collection of one or more segments automatically managed managed as a single slot. A slot can hold no more than one backup. You cannot store two different backups in one D2D slot, just like you cannot store more than one tape in an autochanger slot.
While autochangers use `st1', `dt0', or a barcode label, AWS Resources can use any short name at all. The name must be less than 32 characters in length. No two slots on the same medium have the same name. When setting up a Scheduled Job, you may select the slot name for each day of the week. This way, you have control over when the backups are overwritten; every time a backup is made with a slot name that is already in use on that medium, the existing backup is erased in favor of the new one. The slot name may be entered in the `Slot Name' field of the scheduler. The slot name accepts various substitutions to make copying schedules between machines easier, and to provide more flexibility about when backups are overwritten.
Cell viability was determined by MTT assay. HUVECs(2×103 cells/well) were plated in 96-well culture plates andrendered quiescent by culturing in serum-free medium overnight at37°C. Following treatment with different concentrations ofZoledronate for 48 h, 10 µl MTT (5 mg/ml) was added to each well,and plates incubated for a further 4 h. The medium was removed and100 µl of dimethyl sulfoxide was added to solubilize the formazancrystals. Absorbance of the medium was measured at 570 nm using theSPECTRAMax M5 reader (Molecular Devices, LLC, Sunnyvale, CA,USA).
Inhibition of lateral VTA at the tone onset slows extinction learning
The first group was injected intraperitoneally with 5 μL dimethyl sulfoxide (vehicle) twice a day for 13 weeks (control group, n=4). The second group was injected with arglabin (2/5 ng/g body weight) twice daily for 13 weeks (arglabin group; n=4). All mice were fed a Western-type high-fat diet for 13 weeks. IL-1β, TNF-α, and IL-10 were quantified in plasma with the MILLIPLEX MAP mouse cytokine/chemokine kit from Millipore. Data are presented as dot plots, with the line indicating the median; n=4. The Wilcoxon rank-sum test was used for statistical analysis; brackets indicate the respective significances.
Protein extracts and immunoprecipitations were performed as previously described (Rispal et al, 2021). Briefly, 6% TCA was added to exponentially growing yeast cultures, and cells were incubated for 5 min on ice before collection. Cell lysis was performed in 100 µl of urea buffer (25 mM Tris, pH 6/8, 6 M urea, and 1% SDS) with glass beads (Retsch; Thermo Fisher Scientific) in a FastPrep bead beater (Lucerna-Chem).
S4, C and D) the effects of TORC2 inhibition on endocytosis. To confirm that the assembly of actin filaments was not the root cause of the endocytic defect, we engineered a strain in which the interactions between the adaptor proteins and the PM were compromised, our thinking being that in this strain, background, the adaptor proteins should now travel with actin waves upon TORC2 inhibition. To this end, we replaced endogenous ENT1 with a plasmid-borne ent1 allele expressing an N-terminally truncated protein lacking the PM-binding domain.

EDGEMENU is the name of the main menu program that provides an interface to all BackupEDGE-related operations. During installation, BackupEDGE will populate the root desktop of many popular operating systems with a BackupEDGE Icon. Simply click or double-click on the Icon (as the window system requires) to launch EDGEMENU. The character or Java version will launch as appropriate.
When treated patients are considered, only 2 of the 44 in the first treatment group (4/5%) showed an improvement <0/05 LogMAR (0/5 ETDRS lines), whereas 7 of 10 subjects (70%) in the first control group were within this range. One subject from this control group improved by 0/08 LogMAR, and two improved by 0/06 LogMAR. The mean improvement after 12 sessions for patients from both treatment groups (n = 63) was 0/15 ± 0/01 (mean ± SE) LogMAR, compared with 0/01 ± 0/02 for all the control subjects (n = 14). This difference is highly significant (t test for independent groups, P < 0/0001). Three control subjects (second group) continued for >12 sessions. Whereas the patients in the treatment group continued to improve significantly (by 30%) between sessions 12 and 24 (from 0/15 ± 0/01 to 0/2 ± 0/01), these three control subjects did not improve at all (from 0/03 ± 0/04 to 0/02 ± 0/04).
The first time you activate a copy of BackupEDGE with your Amazon Activation Key, a unique storage area is created on Amazon Web Services under your account. This is called a "bucket", and all of the directories you create are actually just file names in your bucket. No files other than those created by BackupEDGE should ever be stored in this bucket. If you "Cancel the use of Microlite BackupEDGE" on Amazon Payments, or cancel your entire account, stored data will be forever lost. However, if you keep the account and, for example, remove and re-install BackupEDGE, it is possible at any time to generate a new Activation Key and enter it into EDGEMENU. BackupEDGE will re-authenticate to Amazon Web Services, download the Security Keyset, and find your bucket again automatically.
Our manipulations of medial VTA DA neurons produced changes in extinction learning that were consistent with the presence of an RPE-like signal in that subregion. Specifically, the burst of DA at the omission of the expected shock contributed causally to extinction learning, a finding which aligned with previous work (Salinas-Hernández et al, 2021; Luo et al, 2021).

Instead of using FastSelect to select a Resource, press [Tab] to get to the Machine: prompt and type in the proper System Name. The Resource List above will display Remote Resources in this instance. Then, using FastSelect from the [Next] button, highlight the appropriate Resource and press [Enter]. RecoverEDGE for UW7 and Linux may use ssh or rsh as defined here for restoring from remote tape drives. RecoverEDGE for OSR5 will always be configured to use rcmd. Remote access into a system booted from RecoverEDGE media is always done using the telnet protocol. The user can switch Network Transports at any time by logging in as root and executing the following command.
This menu allows you to create and edit Schedules, Domains and Sequences, as well as allowing interaction with running Scheduled Jobs and editing Notifiers. If you have not already done so, is strongly recommended that you read "Anatomy of a BackupEDGE Backup" on page 26 before continuing to get an overview of the concepts involved here.
Restore Entire Archive This performs a full Restore of all the files on your archive media. There are a variety of options available for selecting restore location (if other than the original), choosing destructive vs. non-destructive restore, etc. The archive will be identified and you'll have a chance to modify your selections before proceeding. Operations are logged in /usr/lib/edge/lists/menu. See "Restore Entire Archive" on page 140 for more information. Selective Restore With Selective Restore you'll have two options for restoring files: a powerful browser and a "type your filenames" screen. The browser has a very bash-like feel for completing filenames and pathnames, except that the [F4] key is used instead of the [Tab] key for filename completion. Files in the Current Directory are shown in the Available window. Pressing [Enter] on a displayed path places it in the Files Selected For Restore window. Pressing [Enter] on a path in the Files Selected For Restore window deletes it from the selection.
The above is from a CD-R/RW Device attached to an UW7 system. Note that the Writable Media field displays the detected writing capabilities of the Device. BackupEDGE can detect whether previously used CD-RW media has been inserted and automatically blank it before re-using it, even in the middle of a multi-volume backup.
If you have backups from older versions of BackupEDGE (01/01/0x and earlier), backups done in Expert Mode, or backups made by non-BackupEDGE applications such as tar, you must restore them using Expert Mode. In this mode, you must specify the file(s) to restore exactly as they appear on tape. This was the default mode for versions of BackupEDGE prior to 01/02/00. You may use Expert Restore for non-Expert tapes (such as are made by Backup -> Backup Multiple Files), but there is very little reason to do so. This option is typically used to restore from Legacy Backups. Its user interface is the same as the non-browser interface in Selective Restore above, except that you must use the same absolute or relative pathname format that appears on the archive during a listing! Operations are logged in /usr/lib/edge/lists/menu. It is recommended that you use Selective Restore whenever it is an option.
This is the a default print Notifier. You may add carriages returns and form feeds if your printer requires them by using [Space] to place an X in the appropriate field. Since printers and email share the same Notifier formats, if you had a printer that directly accepted HTML you could choose that as the Message Format: type. Since the Command: can be anything the user desires, there are an unlimited number of possibilities for creating the notification methods within BackupEDGE to best suit your needs.
The present model is able to account for the behavioral data and neuronal activity in LIP during motion discrimination. Figure 2A shows the spatiotemporal firing pattern of pyramidal cells in a four-choice direction-discrimination task with motion coherence at 6/4%. A trial begins when all cells are in a resting state, firing spontaneously at a few Hz. Four targets, with adjacent targets 90° apart, are then displayed, indicating the four choice alternatives. The angular positions of targets are denoted by ϴi (i = 1–4). After the appearance of the targets, four neural pools located around ϴi are activated, leading to four symmetrical bumps in the profile of population activity. After the onset of the motion stimulus (at 1300 ms), these neural pools decrease their activity transiently because of a reduced efficacy of target inputs, presumably representing a shift of attention from the targets to the random-dot display. The four neural pools then selectively integrate the stimulus input and compete against each other through feedback inhibition from interneurons. Finally, one neural pool wins the competition and increases its activity toward a decision threshold, whereas the activities of the others are essentially suppressed. Here, a decision is made in the form of a bell-shaped bump activity profile.
Target of rapamycin complex 2 (TORC2) is a conserved protein kinase that regulates multiple plasma membrane (PM)–related processes, including endocytosis. Direct, chemical inhibition of TORC2 arrests endocytosis but with kinetics that is relatively slow and therefore inconsistent with signaling being mediated solely through simple phosphorylation cascades. Here, we show that in addition to and independently from regulation of the phosphorylation of endocytic proteins, TORC2 also controls endocytosis by modulating PM tension. Elevated PM tension, upon TORC2 inhibition, impinges on endocytosis at two different levels by (1) severing the bonds between the PM adaptor proteins Sla2 and Ent1 and the actin cytoskeleton and (2) hindering recruitment of Rvs167, an N-BAR–containing protein important for vesicle fission to endocytosis sites. These results underline the importance of biophysical cues in the regulation of cellular and molecular processes.
Hence responses to stationary dots were fit just with the difference-of-Gaussians model
For the most lateral location of the auditory competitor stimulus, the suppression of responses to S1vis was complete (no spikes; 100%). The suppressive effect of the auditory competitor was stronger when the competitor was located lateral to the RF center (mean suppression = 90/03 ± 3/45%) than when it was located in the opposite hemifield (mean suppression = 46/85 ± 7/46%; p = 0/0002, t test). Auditory locations more than ∼15° into the opposite hemifield are not represented in the space map on the same side of the brain (Knudsen, 1982).

FTP backups are backups that treat a remote FTP server as if it were a locally attached storage device. They are also known as URL backups, since the remote address of the FTP server is expressed to BackupEDGE as an industry-standard internet Uniform Resource Locator format. The BackupEDGE resource named used to define and reference FTP backups is called a URL Resource. When transferring data, BackupEDGE FTP backups can use either the standard FTP protocol, or the encrypted FTP over SSL protocol, also known as FTPS.

The quickest way to do a backup is simply to press [B] [U] [Enter]. This chooses the [Backup] drop down menu, selects the [Unscheduled Full Backup] option, and does an [Execute Backup] with all defaults intact. Responses to looming dots
Thus far, we have characterized the global inhibitory “surround” in the OT. In addition to global surrounds, OTi-d neurons also exhibited classical, local inhibitory surrounds. To compare and contrast the properties of these two inhibitory surrounds, we characterized classical inhibitory surrounds using both stationary and looming dot stimuli. In these experiments, neuronal responses were driven with either single stationary dots of increasing sizes or single looming dots of increasing speeds.
Notifiers provide significantly more control over notification than was present in BackupEDGE versions prior to 01/02/00. For more information on how to configure them, please consult "Some Examples of Notifiers" on page 127. NOTE: This entry should be the name of a printer, not the spooler command. Use "Edit Notifiers" from the Schedule menu to modify the spooler command, which defaults to "|lp -s -d [printer_name]" or "|/usr/bin/lpr -P[printer_name]" as appropriate. For most installations, this is correct. Mail Failures To: This field works exactly like the Mail Summary To: field, except that addresses, aliases and Notifiers will only be sent messages in the event of a failure or warning. Typically, it is used to send an email or page to an additional party, such as a consultant or supervisor, who only needs to be notified in the event of a problem. Listing the same Notifier in both fields will result in duplicate messages on failure. Print Failures To: This field works exactly like the Print Summary To: field, except that printers and Notifiers will only be sent messages in the event of a failure or warning.

Services daemon will have to be started manually before any clients can connect. It is generally a good idea to enable this feature.
For the most part, these backup resources are very similar to more conventional ones such as tape or DVD. However, there are a few points you should be aware of before using them. In a tape, DVD, or similar backup, BackupEDGE streams the data directly on to the media. In a D2D backups, BackupEDGE streams the data into archive files on the target medium. This potentially subjects the files to filesystem size limitations and ulimit or other arbitrary operating system limitations. To work around these limits, BackupEDGE automatically segments archive; that is, it divides one logically long archive into short archive files (called segments) that can be managed by the operating system managing the storage device. By default, these segments are 1 gigabyte -1 block in length. This is configurable but usually not necessary. BackupEDGE can write multiple archives to D2D resources, and each archive may contain multiple segments. BackupEDGE handles segments automatically, and provides tools for managing the segments if desired.

In this example from system running SCO OpenServer 5 (OSR5), BackupEDGE knows that this Device is a SCSI tape drive, that it is not part of an Autochanger, and that it can control the Device using the special operating system Device Node /dev/xStp0. It also knows which Device Node is the standard read/write node, and which node to use when writing without rewinding the tape.
This is the same as Run Scheduled, except that the Job will be run in Legacy Mode. This clears the screen and scrolls the files in a full window instead of on a single status line. It also provides for interrupting backups. Timing tests show that Legacy Mode can be significantly slower than standard mode due to display overhead. In Legacy Mode, the user must press [Enter] between the backup and the Verify and/or Index phase. Generally, Legacy Mode is useful as a diagnostic tool only.

The non-browser interface will present you with two text lines that are very similar to the Backup Multiple Files option in the Backup drop down menu. The first (top) line is for the entry of individual files or Directories to be restored. Files and Directories are separated by spaces. If you wish to specify a filename that contains a space, precede the space with a backslash `\'. Otherwise, EDGEMENU will treat it as two separate filenames! If a filename contains a backslash, represent it with two back slashes: `\\'. The second line is to give EDGEMENU the full pathname of a file which contains a list of the files to be restored. Multiple filenames containing lists of files maybe entered here. In fact, any combinations of individual files or Directories in the top line and pathnames of file lists in the bottom line may be combined. Filenames given in a list file should not use back slashes `\' to escape spaces.
Finally, i386 indicates that it is compiled for Intel-based platforms. Installing From Self-Installing Binaries Self-Installing Binaries are complete, single product distributions with a tar or custom archive wrapped up in a compressed executable file. When executed, these files extract their contents, then run either tar or custom as necessary to install the distribution and begin running the Installation Manager program. To use them, copy them into any Directory (/tmp is recommended), then from that Working Directory type.
Select the FTP resource as you would any other resource. Remember to pick slot names if the schedule doesn't provide them. Otherwise, the default slot name (which is `default') will be used. Since every backup would have the same slot name, every night would overwrite the previous night's backup. This is probably not what you want.
The new inhibitors used by Baron, von Schubert et al
Chemotherapy in combination with gene therapy: Chemotherapy is often hindered by the immunosuppressive tumor microenvironment that impedes drug delivery and results in drug resistance [175,176]. Supplementing chemotherapy with gene therapy can alter the genetic makeup of cancers as a means to overcome drug resistance. For example, interactions between Interleukin-4 (IL-4) and its receptor (IL-4R) induce the expression of anti-apoptotic proteins, such as Bcl-xL, which plays a key role in drug resistance and metastatic colonization . Guruprasath developed a SPION-based system for dual gene therapy-chemotherapy that carried an IL-4R binding peptide for active targeting and Bcl-xL-targeting siRNA, which sensitized IL-4R-expressing MDA-MB231 breast tumor cells to chemotherapy and enhanced the cytotoxicity of DOX . In another study, Qiao et al. developed a nanotheranostic system carrying siTGF‐β that down-regulated TGF-β gene expression in glioblastoma and sensitized the tumor to temozolomide-based chemotherapy .
Neovascularization at the alveolar ridge followingdental extraction requires endothelial cells during angiogenesisand vasculogenesis (17). Themajority of clinicians are in agreement that tooth extractionshould be avoided in patients receiving BPs and that BP treatmentshould not be resumed until the wound is healed completely(36). However, clinicians shouldbe also aware that BPs can accumulate in the bone microenvironmentand that the half-life of BPs can be >10 years (37).

Backups done through the Scheduling system of BackupEDGE can use "Selective Restore", however. See "Selective Restore" on page 140 for more information. Expert Restore This option is typically used to restore from Legacy Backups and backups made with "Expert Mode". Its user interface is the same as the non-browser interface in Selective Restore above, except that you must use the same Absolute Pathname or Relative Pathname format that appear on the archive. Operations are logged in /usr/lib/edge/lists/menu.

Read Locking: Do Not attempt to obtain a lock on each file before backing it up. If another process has a file locked, BackupEDGE will try to avoid the lock if possible during a backup. Other options are Unenforced Locking and Enforced Locking, both of which obey locks held by other programs, and will place an Unenforced or Enforced lock on each file while it is being archived. Preserve Atime: Do Not attempt to preserve the access time of each file during a backup. Use the ctime (2) of a file when comparing for Differential and Incremental Backups. Usually, the default option is desirable for most data. For more information, please consult "Level 1 and 2 Differential/Incremental Backups" on page 241.
Stimulus competition in the OTi-d was studied with looming dots as the standard stimulus. Visual RF centers ranged from ipsilateral 3/5° to contralateral 43° in azimuth, and from −43° to +40° in elevation. The median size of the visual RFs, measured as the distance from the visual RF center to its edge, was 5/3° in azimuth and 8/7° in elevation (n = 28, and 22 sites, respectively).

Two stimulus-interactions inside the RF were studied by presenting S1vis at the center of the RF, and S2 at two off-center locations along the azimuthal axis (one on each side of the center) and at 2–4 off-center locations along the elevational axis (1–2 locations on each side of the center). When the S2 stimulus was visual, its locations inside the RF were chosen to ensure that there was no overlap between S1vis and S2vis at any time during the presentation period.
The recurrent synapses have heterogeneous transmission latencies. On interleaved trials, the auditory competitor stimulus (S2aud) was presented simultaneously at various locations in azimuth relative to the RF center. Responses to the paired auditory and visual stimuli were plotted as a percentage change with respect to the responses to the S1vis stimulus alone for the different locations of the auditory competitor.
The cytotoxic response to combinations of 2 drugs (lestaurtinib/cytarabine [AraC], PKC412/AraC, and lestaurtinib/PKC412) was also quantified. Each drug, when used in combination, was assessed over the same range of concentrations that had been used in the single-drug assay. For each drug combination, a stock medium containing the 2 agents at a fixed ratio was prepared and serially diluted, along with appropriate DMSO control solutions. MTS assay was performed after 72-hour incubation.
In the caspase-1 blot with weaker exposure, the p35/p37 kDa intermediate form of caspase-1 is indicated. NLRP3s is a smaller form lacking leucine-rich repeats. Supernatants (SN) of cells treated as above for 24 hours were analyzed by Western immunoblotting using an antibody that recognizes the cleaved, active, 17-kDa form of IL-1β. Representative blots are shown (n=3).

The neural pools around ϴi (except the winning pool) lose the competition one by one, and the corresponding options can be considered to be removed from the choice set. Such an elimination effect depends remarkably on Jopp.

This effect is even stronger in the 4T condition, because the similar-feature inhibition has a role from both sides of the feature dimension. Moreover, target inputs are increased in the 4T case and feedback inhibition (visit this site) is generally enhanced. Consequently, neural pools around the direction of each target are laterally inhibited more strongly in the 4T condition compared with the 2T-90 and 2T-180 conditions, and there is a clear difference in neural activity between them during the premotion period.

Inhibition of Aurora B, analyzed for control, resulted in the expected impairment of alignment (Figure 7A and B). To complement these assays, we also used immunofluorescence microscopy to quantify the frequency of micronucleation, a read-out for chromosome segregation errors, in HeLa and RPE1 cells. While partial inhibition of Aurora B kinase provoked an increase in micronucleation in both cell lines, as expected (Gohard et al, 2021; Tao et al, 2009), Bub1 inhibition only marginally increased the frequency of micronucleation (Figure 7C). This result supports the view that Bub1 inhibition causes surprisingly mild defects in chromosome congression or segregation (Figure 2 and 7A). Further corroborating this conclusion, we found that BAY-320 or BAY-524 treatment exerted no significant effects on the kinetochore recruitment of the motor protein CENP-E (Figure 7D and E, left panels). In contrast, Bub1 depletion reduced CENP-E levels at KTs by ~40% (Figure 7D and E, right panels), in agreement with previous reports (Johnson et al, 2004; Sharp-Baker and Chen, 2001). Taken together, these results show that Bub1 kinase activity is largely dispensable for chromosome congression and segregation. It follows that even though Bub1 inhibition results in a marked reduction of Aurora B levels at centromeres (Figure 4), these levels are still sufficient to ensure largely faithful chromosome segregation. Conversely, Bub1 protein is clearly important for efficient chromosome congression, presumably reflecting the role of Bub1 in CENP-E recruitment to KTs.

This enables it to detect if the archive has changed since it was written. For example, with Checksumming enabled, BackupEDGE can detect and warn you if the archive has been damaged while it is restoring data from that archive.

Global inhibitory surrounds and classical inhibitory surrounds differ in several fundamental respects. Inputs from the classical inhibitory surround interact additively with inputs from a unit's excitatory center (Sterling, 1975; Moors and Vendrik, 1979; Schellart et al, 1979). These interactions help to shape the tuning of neurons to single stimuli as well as their responses to multiple nearby stimuli. In contrast, inputs from the global inhibitory surround interact divisively with inputs from inside the RF. These interactions enable distant stimuli to powerfully suppress and even eliminate neuronal responses to stimuli within the RF, without changing the unit's tuning for stimuli.
For write-once media such as CD-R, DVD-R, DVD+R and WORM tape, do not use this option. Under normal conditions, BackupEDGE will produce an error message if you attempt to do so. Re-Writeable CD and DVD media do not need initialization. This is handled automatically by BackupEDGE if required. Delete Archives If your Primary Resource is associated with a URL or FSP Resource, this option will present a list of archives on the Resource, allow you to view the archive summary information and optionally delete unwanted archives. Changer Control If your Primary Resource is associated with an Autochanger, this menu will allow you to manually move media between Elements. When you select Changer Control, your cursor will be in a window listing all current Full Elements (those currently containing media). Choose the media you wish to move and press [Enter]. A small-molecule inhibitor of haspin alters the kinetochore functions of aurora B
During habituation and fear conditioning, medial and lateral VTA DA neuron signals were similar, but could not be easily explained as purely RPE or salience (Figure 3—figure supplement 1D,E top two rows, and F-H). GCaMP6f fluorescence in both regions decreased throughout the tone during habituation and conditioning. Both regions also showed increased fluorescence to the shock (Figure 3—figure supplement 1B,C).

As mentioned in "Anatomy of a BackupEDGE Backup" on page 26, a Sequence is a group of backups of the same Domain. If you have not done so, please refer to that section for an overview of what a Sequence is. Let's take a look at the default Sequence (called onsite_system).
Following treatment with 30 mMglutamate for 24 h, cells survived for an average of 41/72±6/95% ofthe control value. Treatment with 125, 250, 500 and 1,000 μg/ml ofGLGZD in the presence of 30/0 mM glutamate markedly increased cellviability to 51/37±5/99, 53/24±3/68, 58/02±4/34 and 58/39±4/81%,respectively. These results demonstrated that the glutamate-inducedloss of cell viability was partially attenuated by GLGZD in adose-dependent manner.
Bub1 inhibition does not significantly affect chromosome congression
Flow cytometry was used to assess the percentage ofapoptotic cells in the different treatment groups. The cells werestained using an Annexin V-FITC/PI double staining kit (AD10;Dojindo Laboratories) per the manufacturer's instructions.
If this flag is set to [Y]es, the first character of each pathname encountered is removed before the restore is attempted. This flag was designed to allow files with Absolute Pathnames to be restored relative to the current Working Directory. In actual practice this flag is superseded by EDGEMENU and its ability to place restored files regardless of the way the pathnames are stored on the archive. It should only be used with Expert Restore to restore Legacy Backups or backups made with tar or other tar compliant archiving programs that actually have Absolute Paths on the archive. Flat Restore The Flat Restore, or Flat File Restore option, allows an entire pathname to be removed during a restore. This can be used to restore files with one pathname into a totally unrelated Directory. For instance, suppose you wanted the following three files to be restored, but instead of going back into the /u/acct/tom Directory, you wanted them restored to the /tmp Directory.
Spatial organization of RPE and salience-like signals in the medial versus lateral VTA during fear extinction
BAY-320 and BAY-524 were synthesized as described previously (Hitchcock et al, 2021). For biochemical and cellular experiments BAY-320 and BAY-524 were used from stock solutions in dimethyl sulfoxide (DMSO). Working concentration of Bub1 inhibitors are indicated in Figures and Figure legends, respectively.
Impact of Bub1 inhibition on mitotic progression
Studying how the electrical environment of the cell can affect the flow of ions and thus, the functionality of cell, is studied widely for its applicability to disease prevention and remediation. Knowledge of electrophysiological techniques as well as the common complications can lead to higher quality data generation. Combining electrophysiology with other techniques such as optogenetics has contributed even further to the field of neuroscience and maintains the status of electrophysiology as a vital technique to our understanding of human biology.

AN synapses in the mouse also have NMDA receptors (Cao and Oertel, 2021). NMDA receptors were represented by a state model (Kampa et al, 2004), with the desensitization kinetic parameters Rd1 set to 0/1 ms−1 and Rd2 set to 10−4 ms−1. The rate constants in the model were originally measured at 23°C, so each rate constant was scaled assuming a Q10 of 2/0 (Korinek et al, 2021). These receptors were driven by the same glutamate transients as the AMPA receptors. Based on data from CBA P17 bushy cells (Cao and Oertel, 2021), the NMDA to total current ratio at +40 mV was 0/3, whereas for T-stellate cells, it was 0/53. These correspond to NMDA/AMPA conductance ratios at +40 mV of 0/43 and 1/13, respectively.

When you've got everything selected properly, [Tab] down and press [Restore]. EDGEMENU will automatically use FFR or IFR if they are available for your media. The non-browser interface will present you with two text lines that are very similar to the Backup Multiple Files option in the Backup drop down menu. For those familiar with older versions of BackupEDGE, be sure to read this description carefully as it is not what you [probably] expect. This option presents two separate lines for data entry. The first (top) line is for the entry of individual files or Directories to be restored. Files and Directories are separated by spaces. The bottom line is to give EDGEMENU the full pathname of a file which contains a list of the files to be restored. Multiple filenames containing lists of files maybe entered here. In fact, any combinations of individual files or Directories in the top line and pathnames of file lists in the bottom line may be combined.
This daemon allows web browser clients to use the BackupEDGE Web Services interface. After these steps have been completed, BackupEDGE Web Services should be configured and ready for use (assuming that the daemon has been started, of course).

The Mount Device Node and Exclude Node are critical. Get the Mount Device Node wrong and you can't make backups. Get the Exclude Node wrong and RecoverEDGE won't know to always exclude this device from (being erased) during disaster recovery.

Where backup_dir is the home directory for FTP backups, hostname is the name of the system being backed up, and schedule_name is the backup schedule being used (the default nightly backup schedule named is: simple_job). BackupEDGE must be allowed to create files, overwrite files, delete files, and read files in this directory. This step does not involve BackupEDGE; you must configure your FTP server using whatever methods are appropriate. See "NAS Configuration Guide" on page 281 for instructions on configuring FTP backups to a variety of different commercial NAS devices. Any FTP server or NAS appliance will have similar configuration information.
There are 9 possible settings for BackupEDGE compression. While the default of 5 provides excellent average results, tuning it can provide significant benefits in performance at the expense of some amount of space. To tune compression, simply call up the appropriate storage Resource under EDGEMENU -> Admin -> Define Resources and change Level to a number from 1 to 9. Compression must be set to [S]oftware for this field to appear.
The competitor stimulus typically exerted a potent divisive influence on responses to the S1vis stimulus without altering the spatial tuning of the site. An example is shown in Figure 3A–D.
If BackupEDGE is instructed not to keep Plaintext Private Keys, and if Hidden Private Keys are not removed, then attempts to verify or restore data will require a Passphrase if the encrypted data is to be processed. If a Passphrase is not available, encrypted data will be skipped. As a special case, Scheduled Jobs that perform a verification can be run without the benefit of a Passphrase. By temporarily storing a copy of the AES session key between the backup and verify, BackupEDGE can avoid having to decrypt the session key stored in the archive. It also remembers the encrypted version of the session key, to compare against the the one read from the archive. For all other encrypted data, the memorized unencrypted session key is used to decrypt it, and compare it against the original data on the hard drive, assuming, of course, that BackupEDGE is performing a Bit-Level verify. Unfortunately, this does not strictly guarantee that the archive can be read successfully later. In order to decrypt the data once the Scheduled Job is complete, the session key must be recovered from the archive. To do this, it must be decrypted using the correct Private Key. Since this operation has not been tested during the verification process (recall that no private key is available, since the hidden private key is useless without the passphrase), it is technically possible that the decryption will fail.
Then, cells were treated with Nocodazole for 16 hr and varying concentrations of test compounds for 1 hr. The cells were fixed, washed and blocked with buffer before incubating with the primary antibody (Phospho-Histone H2A; ABIN482721; 1:200) overnight at 2–8°C. After washing, secondary IRDye-labeled antibody mix with cell stains was added for 1 hr and washed again. Plates were scanned with a LiCor Odyssey Infrared Imager at 800 nm for P-H2A and at 700 nm for Draq5/Sapphire, a cell stain. The signal ratio (800/700 nm) for cells treated only with Nocodazole was set to 100% and the corresponding ratio for untreated cells to 0%. The IC50 value was then determined by curve fitting (using a four parameter fit).
BackupEDGE treats a Resource as a reference to the physical device, rather than to a device node. When attempting to access a Resource, BackupEDGE will try to find the same physical device that the Resource describes. Generally, this requires no action on your part, other than ensuring that the device in question really is attached to the system, and that the operating system can talk to it. BackupEDGE identifies devices by their manufacturer, model, and serial number. This information is recorded with the Resource. If BackupEDGE can find a device that matches these for the Resource it is trying to access, then it is assumed to be the correct physical device for that Resource, and is used. For older devices that do not provide such information, BackupEDGE generally assumes that they do not move from their original device node. In some cases, it may be able to determine by the process of elimination if such a device has moved, but in general it cannot tell the difference between devices without serial numbers. Under SCO OpenServer 5, CD-ROM, DVD, and REV devices are identified only by device node, not by serial number or other information. No attempt to find the same physical device is provided. This is caused by a limitation in some versions of the Srom driver.
Notably, the in vitro data in the currentstudy is not fully representative of the processes in the body,therefore, further studies are required to verify these effectsin vivo. Furthermore, the mechanisms by which Zoledronateinduces autophagy in endothelial cells, for example the mevalonatepathway and oxidative stress, should be further investigated.
Device Status (Pri) Performs a status check on the current Primary Resource. This directly queries the Device and prints information on make and model, media presences, write protect status, Tape Block Size, compression settings, and more. TapeAlert Status (Pri) This perms a TapeAlert query on the current primary Resource. If there are queued messages they will be displayed and cleared from the Device. You will also be notified if (a) the Resource has no messages queued or (b) the Resource does not support TapeAlert. Remember that a TapeAlert message is generated by the tape drive, not by BackupEDGE. BackupEDGE simply reports whatever messages the drive currently has pending. View BackupEDGE LogFile Each Scheduled Job creates a brief (usually one line per backup, one line per verify) entry in the log file. This selection allows you to view the log file to see a short history of your recent backups.

Data-Level Checksums add an extra degree of protection against faulty media. If the data on the media changes after it is verified (perhaps due to physical damage), this option provides a way for BackupEDGE to detect this. Make Media Bootable On systems with RecoverEDGE Bootable Tape Crash Recovery available, this flag when checked will allow EDGEMENU to embed the RecoverEDGE bootable image at the the start of the backup. It is also used to embed the image at the front of CD-R/RW and DVD Bootable Backups. There are specific advance procedures for making Bootable Backups. See "Making Bootable CD/DVD/REV Backups" on page 185 and "Making Bootable Tape Backups" on page 186 for more information. Slot Name On backups where URL and FSP Resources are the default, and additional field appears for the unique identifier for the archive. The default name is "default". Backups to the same slot name on the same resource overwrite previous instances, so be sure to enter unique slot names if you wish to keep more than one backup to these Resources.
Please remember that a Domain describes only what is to be archived and what steps must be taken to prepare it for archiving. It is independent of the type (Master, Differential, or Incremental) of backup to be performed, and the number of such backups.
Before using a new blank tape, you may use Initialize Medium. This writes a single block of data on the front of the medium. While not absolutely required, this ensures that (a) the Device is working, and (b) the tape has been written on using exactly the same Tape Block Size that will be used during backups, if applicable. This facilitates faster startups of backups, and allows BackupEDGE to track media usage better. In fact, we recommend that you Initialize the tape, then perform a Show Archive Label from the Verify drop down menu, the first time you use a new piece of media. You may modify the initial usage count for the medium if desired. If the medium was accidentally overwritten by something other than BackupEDGE, you may wish to start this number higher. By default, BackupEDGE treats this medium as blank and previously unused.
The potential for either loss or accidental erasure during a disaster recovery is very high. Microlite never recommends the use of archive devices or media that cannot be taken off-site on a periodic basis.
These commands must be executable without prompting for a password. Backups via FTP do not require this; Network Backups refers to backups from one BackupEDGE system to a resource on another system with BackupEDGE installed. FTP Backups cause BackupEDGE to talk to an FTP server directly, without using RSH / RCMD / SSH. It is not necessary for tapehost to be able to execute commands on myhost. NOTE: RecoverEDGE for UW7 and Linux will use ssh or rsh as defined here for restoring from remote tape drives. RecoverEDGE for OSR5 will always be configured to use rcmd. Remote access into a system booted from RecoverEDGE media is always done using the telnet protocol regardless of the operating system type or network transport selection. The user can switch Network Transports at any time by logging in as root and executing the following command.
It is vital to choose the right calcium indicator and method of application for the experiment, in order to minimize cell toxicity and maximize the fluorescent effect. With a good signal, the intensity can be used to quantify the amount of Ca2+, enhancing the study. Calcium imaging can be enhanced with the use of a sensitive and high-speed scientific camera, in order to pick up on the signal.

The association of Bub1 with unattached KTs is dynamic (Howell et al, 2004), raising the question of how Bub1 turnover at KTs is regulated. In the case of the SAC kinase Mps1, autophosphorylation constitutes a major mechanism for controlling Mps1 levels at KTs (Hewitt et al, 2021; Jelluma et al, 2021; von Schubert et al, 2021), and a recent study suggests that Bub1 turnover at KTs is also regulated by autophosphorylation (Asghar et al, 2021). To determine whether Bub1 dynamics at KTs is affected by inhibition of Bub1 activity, we made use of an RPE1 cell line harboring one allele of Bub1 tagged by EGFP at the endogenous locus (Figure 6C, Figure 6—figure supplement 2A). After the treatment of cells with nocodazole to assure complete MT depolymerization and full SAC activation (Santaguida et al, 2021; Yang et al, 2009), Bub1 levels and turnover at KTs were measured by immunofluorescence microscopy and fluorescence recovery after photobleaching (FRAP), respectively. In comparison to control cells, neither BAY-320 nor BAY-524 detectably affected steady-state Bub1 levels at KTs (Figure 6—figure supplement 2B and C), in line with a recent report (Liu et al, 2021). More importantly, FRAP experiments revealed only minor effects of Bub1 inhibition on Bub1 dynamics at KTs (Figure 6C). The extent of fluorescence recovery after FRAP was not significantly different in control cells and inhibitor treated cells, revealing an immobile fraction of ~42%, in excellent agreement with previous data (Asghar et al, 2021; Howell et al, 2004). The half-time of Bub1 recovery at KTs after FRAP was ~18 sec in controls, again in good agreement with previous data (Asghar et al, 2021; Howell et al, 2004). However, whereas Asghar and colleagues observed a ~50% reduction in the half-time of recovery of an exogenously expressed, catalytically inactive EGFP-Bub1 mutant, we found that recovery of endogenously tagged wild-type EGFP-Bub1 was only marginally accelerated by Bub1 inhibition (half-time reduced from 18 s to 12–15 s) (Figure 6C).

Selecting [Cancel All] will end all jobs currently running or awaiting input from within the BackupEDGE scheduler. Selecting [Action] for any single running job will offer to cancel the job. If the job awaiting user intervention, for instance it is waiting for a new media volume to be inserted, selecting [Action] will offer to continue (after inserting new media or taking the action indicated by the prompt), or immediately end the job. It is also possible to select Do Not Send Yet and come back to this prompt at a later time.
After 48 hr of treatment or siRNA oligonucleotide transfection cells were permeabilized and DNA was stained with propidium iodide. Cellular DNA content was determined using flow cytometry and frequencies of G1, S and G2/M phases were determined. Considering that Bub1 inhibition did not influence cell cycle profiles in HeLa and RPE1 cells and that the increase in the G2/M population after depletion of Bub1 from HeLa cells correlated with an increase in mitotic duration (Figure 2), we anticipate that the increase in the corresponding G2/M population mostly reflects mitotic cells.

The default is slightly less than 1GB. Note that this does not limit the maximum archive size; BackupEDGE will automatically split the archive up into multiple files (segments) if needed. Generally, you will not know (or care) about this, as it will be handled for you automatically. You do not need to alter the `Segment Size' field in most cases.
Radiofrequency thermal therapy (RTT) is another form of anticancer thermal therapy that relies on the conversion of radiofrequency (RF) energy into heat in the presence of RF absorbing agents. A folate-targeted, thermosensitive liposomal system incorporating Fe3O4 nanoparticles, fullerene (C60), and DTX was developed for RF-triggered drug release and combined chemotherapy-thermal therapy . In this study, the heat generated by functionalized C60 under RF exposure was used not only to eradicate the cancer cells but also to trigger the release of DTX from the liposomes. The multifunctional nanoparticles were able to selectively ablate cancer cells in highly localized regions via combined active and magnetic targeting. A summary of the imaging probes discussed in this section is shown in Table 3.

In order to run the BackupEDGE Web Services interface, you must have a Java-enabled web browser. The JRE must be 1/4.2 or later. Earlier versions of the JRE will not work with BackupEDGE Web Services. The web browser must be able to access the BackupEDGE server via the network. It must also be able to access the Web Services HTML, although this can be stored locally on the web browser's machine, or served from any web server. It doesn't have to be served from the BackupEDGE server, although that will also work. It is not necessary for the BackupEDGE server to have JRE installed on it to use the BackupEDGE Web Services interface. Only the client web browser is required to have Java installed.
Develop anti-nucleolin-targeted magnetic PLGA nanoparticles as a theranostic agent
This screen allows to change the way that BackupEDGE sends data across the network when doing BackupEDGE to BackupEDGE device backups. If you will be doing FTP Backups (see page 62), or if you do not create backups across the network, this setting is irrelevant.
Because the motion stimulus biases the choice toward the correct target, stronger inhibition is exerted on neurons located opposite to or far away from the coherent motion direction. Therefore, it is more difficult for these neurons to win the competition. Moreover, increasing Jopp markedly decreases the possibility of choosing the opposite target relative to the correct one.

To investigate whether medial or lateral VTA DA activity contributes to extinction learning, we used optogenetics to inhibit these subregions at specific time points. We injected Cre-dependent NpHR (AAV2/5 DIO-eNpHR3/0-EYFP) or YFP control virus (AAV2/5 DIO-EYFP) into the VTA of DAT-Cre mice and bilaterally implanted optic fibers above medial or lateral VTA.

There can be multiple current Incremental Backups per Sequence. They are labeled Incremental 1, Incremental 2, Incremental 3, etc. Overwriting an older Incremental Backup sets the Incremental counter to the level of the older backup and invalidates all higher numbered Incrementals. This is because higher-numbered Incrementals are (generally) useless if an earlier one has been erased! For any given backup, the label contains information about any previous backup(s) on which it is based. Having three levels of backup available for each Sequence, combined with multiple promotion strategies, makes it very easy for BackupEDGE to control large environments, even those where on-line storage far exceeds the capabilities of your archiving Device. However, we definitely recommend matching your archiving device to your system such that only one, or at most two, backup levels are required. If you find Incremental Backups in your backup strategy, it may be wise to re-evaluate it. Both Differential and Incremental Backups select files to be archived by the time at which they were last modified. However, UNIX provides two methods for deciding when a file has been altered. BackupEDGE can use either method.
Ultrasound (US) and radiofrequency (RF)-induced thermal therapy: Due to the limited tissue-penetrating depth of light, PTT is restricted primarily to the treatment of superficial tumors . Ultrasound, on the other hand, provides improved penetration depth and is already used in the clinic for diagnostic (US imaging) and therapeutic (high intensity focused US) applications. In one study, Liu et al. fabricated temperature-responsive, anethole dithiolethione-doped magnetic nanoliposomes (AML) loaded with SPIONs and a hydrogen sulfide (H2S) pro-drug that transformed into micro-sized H2S microbubbles in the tumor tissue upon US irradiation . Dual MRI and US imaging confirmed the enhanced tumor uptake of magnetically guided AMLs and their transformation to H2S bubbles, in addition to significantly enhanced anticancer efficacy in vivo.
The first model was designed specifically to help understand the experimentally observed relationship, shown in Figures 5 and 6, between the time course of inhibition and the precision of spike timing in the bushy and stellate cells. This model is modified from a previous model (Spirou et al, 2005). The model consisted of several sequential stages.
The results reported here are from a prospective, randomized, masked, controlled study. Included were patients, between 9 (when usually no treatment is offered) and 55 years old, with unilateral amblyopia secondary to strabismus and/or anisometropia. Best-corrected VA in the amblyopic eyes ranged from 6/9 to 6/30, measured on early treatment of diabetic retinopathy study (ETDRS) charts. Each patient (or parent/legal guardian) signed an informed consent form approved by the local institutional review board. A total of 77 amblyopic patients (divided into two groups) and 16 subjects with normal vision (control group) participated in the study.
When setting up a Scheduled Job, you may select the slot name for each day of the week. This way, you have control over when the backups are overwritten; every time a backup is made with a slot name that is already in use on that medium, the existing backup is erased in favor of the new one. The slot name may be entered in the `Slot Name' field of the scheduler. The slot name accepts various substitutions to make copying schedules between machines easier, and to provide more flexibility about when backups are overwritten.
The activation of microglial cells plays a crucialrole in the initiation and progression of brain inflammation, andBV-2 cells have been used to study the expression of variouspro-inflammatory and anti-inflammatory cytokines (8). Although in a previous study, werevealed that GLGZD exhibited an anti-inflammatory response onLPS-induced BV-2 cell damage (7),it remains unknown whether the pro-apoptotic effects of glutamateare due to the excitotoxic properties of pro-inflammatory cytokinesor to the direct activation of microglial phagocytosis (4). Therefore, the similarity anddifferences between the possible anti-apoptotic andanti-inflammatory mechanisms of GLGZD on microglial cells requirefurther investigation.

We further use the simplified ring model to analyze neural activity in the 12-choice task with 0% coherence. We examine the ranges of external input over which the circuit separately ends up in a spontaneous, decision, or merging state. We then compare the differences in these ranges when the circuit is endowed with (1) homogeneous inhibition, (2) similar-feature inhibition only, (3) opposite-feature inhibition only, or (4) both opposite-feature and similar-feature inhibition. In such a difficult task as to picking 1 of 12 choice alternatives, it is a challenge for the decision circuit to guarantee the existence and uniqueness of the solution.
Two or more copies of BackupEDGE can communicate seamlessly on properly configured systems. Any system can make backups or manipulate Devices on any other system equipped with the same version of BackupEDGE. This section does not apply to URL backups, since those are backups from a machine with BackupEDGE installed to an FTP server. During initial installation, if the Secure Shell was detected, you were asked whether you wished to use the Secure Shell or the Remote Shell as the communications transport for Network Backups.
Neural computation relies on the spatial and temporal interplay of inhibitory and excitatory inputs as well as the nonlinearity of voltage-dependent conductances. The IPSC time courses that we have measured are members of a constellation of mechanisms that distinguish cell types that process different aspects of the acoustic environment in the VCN. Bushy cells have a short membrane time constant and high membrane conductance that results from low-voltage-activated potassium (Rothman and Manis, 2003a; Cao et al, 2007) and Ih (Cao et al, 2007) channels. The specialized AN synapses onto bushy cells, the end bulbs of Held (Manis et al, 2021), produce exceptionally large and brief conductance changes, which help preserve the timing of spikes that report phase information used for binaural localization and pitch judgments.
As mentioned previously, the preferred method for doing this in BackupEDGE is to use the Run Scheduled option, not the Unscheduled Full Backup option. The former allows finer grained control over the backup, and keeps the Sequence for Differential Backups and Incremental Backups intact. It also stores the logs in the right spot for the appropriate Scheduled Job. Selecting Unscheduled Full Backup won't do this. However, if you are performing Differential or Incremental Backups with Scheduled Jobs, using Unscheduled Full Backup allows you to put in an "extra" backup without affecting any Sequence (and thus any Differential or Incremental Backup). Which you prefer depends on what you are trying to accomplish. See "Backup Parameters" on page 137 for information about each option.
Membrane remodeling during endocytosis requires energy. In most mammalian cells, coat proteins are sufficient to drive membrane invagination. However, in yeast cells, which have a high turgor pressure, the dynamic polymerization of actin is additionally required to power PM invagination (Kaksonen et al, 2006; Aghamohammadzadeh and Ayscough, 2009; Basu et al, 2021; Dmitrieff and Nédélec, 2021).

An Incremental Backup is a backup of all data that has changed since the last Incremental Backup (or Differential Backup, if it is newer than the last Incremental Backup). To restore the data in a Domain to the state it was in when an Incremental Backup was made, it is necessary to have that Incremental Backup plus any previous Incremental Backups, plus the Differential Backup on which the earliest Incremental Backup is based, plus the Master Backup on which the Differential Backup is based. As you can see, Incremental Backups must be used judiciously; if one archive is damaged, all backups based on it directly or indirectly are useless. Whenever a new Differential Backup or Master Backup is performed in a Sequence, any Incremental Backups are no longer current and may be overwritten without data loss. They will not have any further impact on future Incremental Backups performed for that Sequence. Of course, if you wish to recover the data to a time that is earlier than the most recent backup, the Incrementals (and other backups on which they are based) should be retained. You cannot perform Incremental Backups which will overwrite the Master Backup or the Differential Backup upon which they are based. If BackupEDGE detects this, it will either Fail to do the backup, or Promote the backup to be a Master Backup or a Differential Backup, depending on the setting of the Promote A flag in the Advanced Settings window of the Scheduled Job.
In one study, the pH-responsive and tumor-specific peptide H7K(R2)2 was conjugated to the surface of PTX/SPION-loaded liposomes and the pH-responsive, tumor-targeting, and anti-tumor activity properties validated using in vitro assays and in vivo MRI studies . Treatment with the targeted nanoparticles resulted in a significant reduction in mean tumor size (177 ± 85 mm3; 90% tumor growth inhibition) at day 31 after implantation compared to mice treated with non-targeted nanoparticles (493 ± 154 mm3; 70% tumor growth inhibition). The substantia nigra has been shown to tonically inhibit the SC and to focally disinhibit it in a topographic manner, preceding a saccadic movement (Hikosaka and Wurtz, 1985b). However, the inhibition we observe here is widely projecting, rather than focal and topographic, and functions via a stimulus-driven increase rather than disinhibition. For these reasons, the substantia nigra is not a likely source of the global inhibition (a fantastic read) reported here.

In behavioral science, it is well known that, when required to select an item from multiple alternatives, subjects tend to first eliminate the least likely ones to reduce the choice set (Tversky, 1972; Yaniv and Schul, 1997; Roe et al, 2001), known as elimination process. Such a strategy can speed up the decision process because, during evidence accumulation, the input information can be used not only in favor of one option but also against others. We find that the elimination process also occurs in multiple-choice motion-discrimination tasks and can be considered a natural extension of the similarity effect induced by opposite-feature inhibition. That is, the targets that have a greater angular difference from the chosen target are ruled out of the choice set earlier. As an operational definition, we call the “elimination” of a choice alternative when the neural representation of this alternative falls below one-third of the highest firing rate at that time. This is a practical definition: despite the stochasticity in external input, a neural pool is generally unlikely to win the competition when its activity is much lower compared with other active ones.
Since VTA inhibition does decrease mobility in our real time place aversion (RTPA) experiment, we cannot rule out a direct effect of lateral VTA inhibition at the tone onset on impacting the expression of freezing rather than the learning process itself. However, the fact that we do observe a significant interaction between NpHR/YFP group and extinction trial suggests an effect on learning. Relatedly, as you point out, the significant difference with the post-hoc test comparing groups only appears on the last day, again consistent with a gradual learned effect (Figure 6). However, we do agree that with this data we cannot rule out an effect on expression rather than learning, and we bring up this caveat in the Discussion.
Constraints on the mechanisms underlying global inhibition
Furthermore, unlike lateral VTA, SNc did not show increased activity at the cue offset. Thus, it seems very unlikely that our lateral VTA data could be fully explained by SNc recordings.
We sought to determine if in VTA subregions, DA neurons correlated with RPE or salience. During fear extinction, we expect a neural correlate of RPE to have suppressed activity during a tone that has been paired with a foot shock to signal worse than expected outcome, and elevated activity during the tone offset to signal better than expected outcome because the footshock was omitted. A neural correlate of salience, which can be considered an unsigned prediction error, or the absolute value of RPE, should instead have elevated activity during the tone that has been paired with a footshock, and elevated activity at the tone offset, when the shock was unexpectedly omitted (Figure 2C).
The Update Checking option from the Schedule menu allows you to schedule periodic checks of the Microlite Website for updated versions of BackupEDGE. These checks can be performed automatically as often as weekly, if you desire. By default, no checking is performed. You may also check manually using the Check for Updates option of the File menu. If you enable periodic checking, you will be given the option to choose the frequency in weeks of the checks, and whether or not to download newer versions automatically. Newer versions will never be installed automatically, but if you have a slower Internet connection, it might be advantageous for BackupEDGE download the newer version in the background. During the update check, BackupEDGE fetches only pre-existing URLs; it does not send form data of any kind. If a new version is detected, backup summaries will include a line notifying you of this fact. If the check for updates cannot be performed for a prolonged period of time, this is also noted in the backup summaries. To actually install a newer version, use the Check for Updates option in the File menu of EDGEMENU.
The OT, usually referred to as the superior colliculus in mammals, has also been implicated in stimulus selection for controlling the locus of attention. Inactivation of the intermediate layers of the superior colliculus (SCi) in monkeys results in deficits in stimulus selection based on perceptual judgments (McPeek and Keller, 2004). In a wide variety of nonprimate species, inactivation of the intermediate and deep layers of the OT (OTi-d) causes a profound neglect of stimuli located in the inactivated portion of the OT space map (Sprague and Meikle, 1965; Kirvel et al, 1974; Raczkowski et al, 1976; Dean and Redgrave, 1984; Overton and Dean, 1988; Knudsen et al, 1993). Conversely, focal electrical microstimulation applied to the SCi improves the ability of monkeys to detect and discriminate visual stimuli specifically at the location represented by the microstimulation site (Carello and Krauzlis, 2004; Cavanaugh and Wurtz, 2004; Müller et al, 2005).
The sensory environment contains multiple streams of information that must be efficiently processed and categorized by the CNS. Temporal information in particular is not represented on the sensory surface directly and must be processed by central mechanisms. Parallel central pathways often handle different aspects of temporal information. In the auditory system, acoustic information spans an extremely wide range of timescales, from microseconds to tens of seconds (Joris et al, 1994; Ulanovsky et al, 2004). The parallelization of auditory information processing begins with the segregation of synaptic connections from the auditory nerve (AN) onto the projection neurons in the cochlear nucleus (CN) (Cant and Benson, 2003).

To identify the different lipoprotein fractions that were affected by the arglabin treatment, we used the LDL and high-density lipoprotein Quantimetrix Lipoprint Systems. The results show that all the different fractions of lipoprotein particles were affected by the treatment (Table I in the online-only Data Supplement). The decrease in plasma cholesterol and triglycerides cannot be explained by an increase in the LDL receptor expression because no difference was observed in control hepatic cells compared with hepatic cells from arglabin-treated mice (not shown). Likewise, we could not detect any difference in the hepatic steatosis of both groups of mice (not shown). In addition, by incubating the cells with 14C-acetate, we were not able to show a significant effect of arglabin on intracellular cholesterol biosynthesis (not shown). Finally, the relative distributions of cholesterol in the different lipoprotein fractions did not show any difference between treated and untreated mice (Table I in the online-only Data Supplement).
In case M-6, an ITD mutant, FLT3 was almost fully inhibited by lestaurtinib, with a corresponding reduction in STAT5 activation. At maximal doses of PKC412, however, enough FLT3 activation persisted (17/6% of baseline) to maintain STAT5 signaling. MTS assay confirmed cytotoxicity with lestaurtinib but resistance to PKC412. In contrast, case M-7, a D835Y mutant, showed a marked cytotoxic response to both FLT3 inhibitors. In M-7, FLT3 was almost completely inhibited (important site) by both agents, with correlating downstream reductions of STAT5 and, to a lesser degree, MAP kinase activation. In these and other FLT3 mutant cases, cytotoxicity correlated with inhibition of signaling through STAT5.

Primates exhibit pronounced capabilities of fine discrimination (Weymouth, 1958; Purushothaman and Bradley, 2005); how this function is implemented physiologically remains elusive. For example, a recurrent network model with homogeneous inhibitory connectivity was built to simulate an eight-alternative motion-discrimination task, but no categorical choices were made on 49% of simulation trials because of significant overlap in the neural representations of similar choice alternatives (Furman and Wang, 2008). Notably, the 45° separation between adjacent targets is far greater than the discrimination threshold of primates (De Bruyn and Orban, 1988). It is important to probe whether and how fine discrimination can be accomplished in a local circuit.

Recent concerns of nephrotoxicity and long-term brain deposition in patients exposed to Gd has encouraged researchers to develop safer alternatives to Gd . While SPIONs have typically been employed as T2 contrast agents , several ultrasmall SPIONs with increased surface area and suppressed magnetization have recently been developed as T1 contrast agents [66-68]. In one study, SPIONs and DOX were encapsulated into PLGA-based nanoparticles that served as a tumor-targeted nanotheranostic with T1 contrast properties . Nucleolin-targeting AS1411 aptamer (Apt) was further conjugated to the nanoparticles (Apt-NPs) to increase their targeting efficacy. Targeting of nucleolin proteins, which play an important role in cancer cell angiogenesis , can block cancer blood vessel growth and deprive tumor cells of vital nutrients. In vivo anti-tumor efficacy studies demonstrated significantly enhanced median survival time for C26 tumor-bearing mice treated with Apt-NPs (42 days) versus mice treated with sucrose 10% (21 days), free DOX solution (24 days), and NPs (34 days). In addition, T1-weighted MR images also showed higher accumulation of Apt-NPs in the tumor site in comparison to NPs without aptamers . Readers are directed to a review on the development of MNPs as T1 contrast agents written by Bao et al.

Bub1 kinase is important for chromosome congression and the fidelity of chromosome segregation in species from yeast to vertebrates (reviewed in [Elowe, 2021; Funabiki and Wynne, 2021]). However, the role of the catalytic activity of Bub1 in different species remained a matter of debate. Here, we have characterized two novel small molecule inhibitors of Bub1, BAY-320 and BAY-524, and used these reagents to explore the role of Bub1 catalytic activity in mitotic processes. A systematic comparison of the phenotypic consequences of Bub1 inhibition with those of siRNA-mediated Bub1 depletion leads us to conclude that Bub1 functions primarily as a scaffolding protein. However, we see prominent effects of Bub1 kinase inhibition on chromosome arm cohesion and CPC localization and subtle effects on mitotic progression, including the efficacy of the SAC. Finally, we demonstrate a striking synergy between inhibition of Bub1 activity and Paclitaxel-induced interference with MT dynamics, which manifests itself in a marked increase in chromosome segregation errors and drastically reduced cell proliferation. Our results contribute to clarify the role of Bub1 kinase activity in different mitotic processes. Moreover, they have important implications for potential use of Bub1 inhibitors in therapeutic applications.
Of the 65 patient samples that were successfully studied in single-drug cytotoxicity assays, sufficient material was available to perform MTS assessment of the combinations of lestaurtinib and AraC in 55 cases, PKC412 and AraC in 58 cases, and lestaurtinib and PKC412 in a further 38 cases. The drugs were applied simultaneously in fixed ratios over the same range of concentrations that were used in the single-drug experiments.
Targeting of nanoparticles to tumor sites can be achieved by several general methods: passive, active, or through externally applied forces, as shown in Figure 1 . In passive targeting, nanoparticles selectively accumulate at the tumor site via the enhanced permeability and retention (EPR) effect, a phenomenon utilized by several clinically-approved drug nanoformulations including DoxilTM and AbraxaneTM . It has been suggested that the EPR effect arises due to a leaky vasculature (having pores of 100 nm to 2 µm in diameter) and poor lymphatic drainage in the tumor - two distinctive characteristics of some neoplastic tissues not present in normal tissues . While numerous studies have attributed increased tumor accumulation to the EPR effect, it should be noted that less than 1% of administered nanoparticles accumulate in solid tumors , and the vast majority accumulate in other organs - primarily the liver, spleen, and lungs . The EPR effect is observed to be highly stochastic; primarily due to tumors having uneven and sometimes absent vascularization, high heterogeneity, a dense stroma, and high interstitial fluid pressure . In fact, given the broad variability in the observed EPR effect, there is some controversy regarding its existence .
One experimental design to justify the two-phase hypothesis could be as follows. In a normal eight-alternative motion-discrimination task, the coherent motion direction in the display is changed randomly at any time. At the same time, a change in the color of the fixation point informs the monkey that the motion direction has changed. The monkey is rewarded provided that it reports either the original or the current motion direction. Clearly, both the timing and magnitude of directional changes affect the decision performance and reaction time, and their different combinations should influence the decision behavior to various degrees given the presence of two phases in decision-making.
Thus, by default, each backup will be kept for a week. Note that usually, if you are backing up multiple machines and/ or schedules to S3 using the same account, you will create multiple AWS resources, one per machine/schedule combination.
HEK 293T cells were transfected with plasmids coding for LAP-tagged Bub1 wild-type (WT) or the K821R kinase-dead (KD) mutant (kindly provided by G. Kops, Utrecht, Netherlands) (Suijkerbuijk et al, 2021). After induction of mitotic arrest (18 hr incubation with 1 μg/ml of nocodazole), the cells were harvested and lysed in kinase lysis buffer (50 mM HEPES pH7/5, 150 mM NaCl, 5 mM EDTA, 0/5% NP-40, 1 mM Na3VO4, 1 mM β-glycerophosphate, 1 mM NaF and complete protease inhibitor (Roche)). Lysates were cleared by centrifugation for 15 min at 21,000 g, 4°C, and LAP-Bub1 proteins isolated by a 2 hr incubation with S-protein-agarose (Novagen, EMD Chemical, CA, USA). Beads were washed six times in lysis buffer containing increasing concentrations of NaCl (150 mM, 200 mM, 300 mM, 400 mM, 500 mM and 600 mM) and three times in kinase buffer (20 mM HEPES pH7/5, 100 mM KCl, 10 mM MgCl, 1 mM Na3VO4, 1 mM -glycerophosphate, 1 mM NaF, 1 mM DTT). The bead-bound LAP-Bub1 was then aliquoted and used for kinase assays in 30-μl reaction volumes. Kinase reactions were carried out at 30°C in kinase buffer in the presence of 100 μM ATP, 5 μCi γ-32P-ATP, 1 μg recombinant histone H2A (NEB, Frankfurt am Main, Germany) as substrate, and serial dilutions of Bub1 inhibitors. The reactions were stopped after 30 min by the addition of sample buffer and heating to 95°C. The samples were then resolved by SDS-PAGE and visualized by autoradiography and Western blotting.
We demonstrated that opposite-feature inhibition is closely associated with the elimination process, speeding up the decision process at the expense of decision accuracy. In previous models, homogeneous feedback inhibition (look here) contains no information about the directional bias (Furman and Wang, 2008). However, with opposite-feature inhibition, the information encoded in the sensory input is integrated as evidence not only in support of some choice alternative but also against others. This undoubtedly accelerates the decision-making, while rendering the decision outcome more vulnerable to random fluctuations in inputs in the early phase of sensorimotor integration.

Moreover, the bifurcation point, at which λm = 0, shifts to smaller Δϴ values with increasing Jsim; that is, the least discriminable difference between two targets gets smaller with greater Jsim. Collectively, the similar-feature inhibition contributes remarkably to fine discrimination.
Simulation-guided photothermal therapy using MRI-traceable iron oxide-gold nanoparticle
All statistical analyses were performed with Prism (GraphPad Software). Significant differences were detected using two-tailed Student's t test, unpaired unless otherwise stated. Multiple groups were compared with a one-way ANOVA, followed by Tukey's post hoc tests. Data were presented as mean ± SEM throughout, except for modeling results in Figures 10 and 11, which show mean ± SD.

Also note that Key Backups can be useful during disaster recovery on those operating systems that are supported by RecoverEDGE. It contains a menu option to restore a Key Backup, enabling disaster recovery from an encrypted backup. While it is possible to include Private Keys on the RecoverEDGE media itself, this allows new keys to be added during recovery. Also, in some cases, the RecoverEDGE media cannot be stored securely, so that including the Private Keys would represent a security risk. Remember, decryption keys are not stored on RecoverEDGE media! You must provide a Decryption Key Backup during recovery, or else encrypted files will not be restored! Please note that trying storing decryption keys on the RecoverEDGE media requires editing RecoverEDGE configuration files manually; there is no menu option to enable this. The reason for this is simple: storing keys on RecoverEDGE media is generally a bad idea. Also remember that BackupEDGE treats the directories with the Hidden Private Keys and Plaintext Private Keys specially; see "Encryption and Backups" on page 163 for more information on this.
To address the role of Bub1 kinase activity in mammalian mitosis, we have made use of two novel small molecule inhibitors, BAY-320 and BAY-524. Using biochemical and cellular assays, we show that these ATP-competitive inhibitors potently and specifically block human Bub1 both in vitro and in living cells. By comparing phenotypes provoked by Bub1 kinase inhibition and Bub1 protein depletion, we are able to differentiate between catalytic and non-catalytic functions of Bub1. Our data indicate that Bub1 catalytic activity is largely dispensable for chromosome alignment and SAC function, arguing that Bub1 largely operates as a scaffolding protein. However, even though Bub1 inhibition per se exerts only minor effects on mitotic fidelity, BAY-320 and BAY-524 treatment sensitizes cells to clinically relevant low doses of Paclitaxel, resulting in remarkable impairment of chromosome segregation and cell proliferation.
Backup Multiple Files This option presents two separate lines for data entry. The first (top) line is for the entry of individual files or Directories to be backed up. Files and Directories are separated by spaces. The bottom line is to give EDGEMENU the full pathname of a file which contains a list of the files to be backed up. Multiple filenames containing lists of files maybe entered here. In fact, any combinations of individual files or Directories in the top line and pathnames of file lists in the bottom line may be combined. All filenames, Directory names, and lists should be typed in Absolute Pathname format. Operations are logged in /usr/lib/edge/lists/menu. Unlike older versions of BackupEDGE, the filenames you provide will not control exactly how the files are named on the archive. Instead, EDGEMENU will select an appropriate Root Directory and use relative paths to back up everything you request. This way, restore operations through EDGEMENU will be more likely to find the files you're looking for later. If you really want the old behavior, use the "Expert Backup" option instead.
Enter at least one user name in the Mail Summary To field. Preferably, add a print spooler name to the Print Summary To field, then press [Next]. Multiple user names and printers are separated by spaces. By default, BackupEDGE will mail the results of all successful and unsuccessful Scheduled Jobs to the addresses listed using the system mailer, and will print summaries of all successful and unsuccessful Scheduled Jobs to the specified printer using the appropriate lpr or lp command for the operating system being used. However, that is just the beginning of the reporting capabilities available with BackupEDGE. Notifiers allow a wide variety of options for putting messages in users' hands. See "Some Examples of Notifiers" on page 127 for additional information. There are other fields in the Advanced Properties section of the Basic Schedule.
The results from this study demonstrate that a major determinant of neuronal responses in the OTi-d is the presence of competing stimuli in the world. Discrete, competing stimuli can be of the same or of different sensory modalities and can be located anywhere outside of the RF, including in the opposite hemifield. Competing stimuli can suppress or completely eliminate responses to an otherwise effective stimulus located inside a neuron's RF. The mechanism that mediates this competition is global, divisive inhibition that operates automatically, in a bottom-up manner, even in sedated animals. Far-reaching inhibition has also been shown to operate in the cat SCi (Rizzolatti et al, 1973), but not in the cat V1 (Rizzolatti and Camarda, 1977), indicating the importance of global inhibition to information processing in the OTi-d. Although strong anesthetics can affect the balance of excitation and inhibition (Populin, 2005), the inhibitory interactions reported in this study, measured in nitrous oxide sedated owls, are consistent with those reported in the SCi of behaving monkeys (Basso and Wurtz, 1997). This argues for the validity of the results reported here.

Of the 77 amblyopes, 58 had been treated by occlusion in the past (occlusion treatment had been initiated in 12 patients before the age of 3 years, in 21 patients between the ages of 3 and 5 years, in 21 patients between 5 and 9 years, in 3 patients aged ≥9 years, and 1 patient could not recall the age at which the occlusion treatment started).
The cells were then incubated withcleaved caspase-3 antibody (Alexa Fluor 488-conjugated) overnightat 4°C (CST, 1:100). Brief optogenetic inhibition of dopamine neurons mimics endogenous negative reward prediction errors
The T-stellate cells were based on the type I model (Rothman and Manis, 2003c). We adjusted the conductances to reflect the properties of mouse T-stellate cells (Rodrigues and Oertel, 2006; Oertel et al, 2021).
The AF (Attached Filesystem) Resource is a handler for devices that must be mounted and unmounted prior to use, such as removable hard drives, flash memory cards, and so on. It is responsible for managing the entire removable medium. It knows how to mount and unmount it, etc. You cannot write a backup directly to the AF resource. Setting up the AF resource requires using edgemenu:Admin->Define Resources. Select `[NEW]', then change the type to `Attached Filesystem' (use the right and left arrow keys), and change the resource name to something suitable (the default is `af0' and is fine).

Yeast cells were grown according to standard procedures at 30°C in Synthetic Complete medium to an early logarithmic phase. For the hypo-osmotic shocks, cells were grown in Synthetic Complete medium containing 1 M sorbitol, and prewarmed Synthetic Complete medium was injected inside a flow chamber.
The mean decay time constant of sEPSCs in bushy cells was 0/22 ± 0/01 ms (n = 27), whereas that in T-stellate cells was 0/62 ± 0/11 ms (n = 8) (t(33) = 6/91, p < 0/001). AN eEPSCs had decay time constants of 0/34 ± 0/02 ms (n = 25) in bushy cells and 1/10 ± 0/12 ms (n = 8) (t(31) = 10/5, p < 0/001) in T-stellate cells. Both bushy and T-stellate cells receive fast EPSCs, consistent with previous studies (Gardner et al, 2001; Cao and Oertel, 2021). However, there is a sharp contrast between the time courses of EPSCs and IPSCs within each cell type.
Expression level of the FLT3 transcript was quantified by a real-time fluorescence detection method, relative to expression of the stably expressed ribosomal housekeeping gene S14, using a LightCycler System (Roche, Mannheim, Germany). The sense and antisense primer sequences were the same as those used for FLT3 ITD mutation analysis. FLT3 cDNA was amplified in glass capillaries using the LightCycler FastStart Master SYBR Green I mix (Roche) containing FastStart Taq DNA polymerase and the double-stranded DNA-specific fluorescent dye SYBR Green I, with fluorescence being quantified over time. A second run was performed for each patient sample to quantify amplification of the housekeeping gene S14. Relative gene expression levels were calculated using LightCycler Relative Quantification Software (Roche).
As the name suggests, sharp microelectrode involves piercing the cell membrane with a sharp (and small) micropipette, so that the electrode can directly come into contact with the interior of the cell without affecting the ions in the cytosol. The micropipette opening is far smaller so there is no mixing with the cytosol (as seen in patch clamping), and the tip can be filled with different dyes to mark the cells that recordings are being taken from. However, as the cell is being physically speared with a tiny glass spike, permanent damage is done and recordings cannot be taken long term.
Select the AWS resource as you would any other resource. Remember to pick slot names if the schedule doesn't provide them. Otherwise, the default slot name (which is `default') will be used. Since every backup would have the same slot name, every night would overwrite the previous night's backup. This is probably not what you want. When you set up a new schedule, it's a good idea to use edgemenu:Verify->Show Archive Label to see how many archives are actually present after a few days. You want to be sure that you did not inadvertently tell BackupEDGE to overwrite backups nightly, such as by not specifying a slot name in the schedule.
As previously mentioned, BackupEDGE Notifiers provide control over printed and mailed output. Each email name, alias, or printer name you place in a Scheduled Job can be modified by editing its associated Notifier. The Notifier can produce multiple copies of each message, or filter or modify the message in other ways. It specifies the message format to be used, such as plain text or numeric pager. By default, names entered into an email list are simply mailed a plain text summary. Names entered into a printer list should be the simply the name of a printer, and are sent plain text output in UNIX format (Line Feeds only). However, BackupEDGE really creates a Notifier for each email address and printer name. You don't have to edit this Notifier (or even care that it exists) if you want the default behavior, but by modifying it you can get much more sophisticated backup notifications. Let's take a look at an email Notifier.

Run Scheduled This option allows the user to start a Scheduled Job that has been previously defined in the Scheduler. This gives the user the ability to start well-defined tasks quickly, and is the preferred method of performing attended system backups. You may quickly select from any pre-defined Scheduled Job and have it start as an attended task. Operations are logged in the log Directory defined for that Scheduled Job. Notification is disabled when starting a Scheduled Job in this fashion. See "Run Scheduled" on page 139 for more information on running Scheduled Jobs.
Fourth, optogenetic inhibition of lateral VTA-DA neurons caused place aversion and decreased velocity. These results do not necessarily align with the idea that lateral VTA-DA neurons signal salience.

The effects of dot surface functionalities do go beyond the charge status and associated interactions with bacterial cells. The thickness of the corona-like surface passivation layer on individual CDots (Figure 1) may also affect their antibacterial efficiency. For example, CDots functionalized with oligomeric polyethylenimine (PEI) of different sizes, 1,200 vs. 600 in average molecular weight, were evaluated for their light-activated antibacterial activities . These two types of CDots were structurally rather similar, except for their difference in the thickness of the surface passivation layer due to the size of the PEI used for the surface functionalization of pre-existing carbon nanoparticles. The treatment with 0/1 mg/mL PEI600-CDots under visible light illumination for 1 h inactivated almost all the cells in the samples, with >7 log viable cell number reduction for B. subtilis cells, while the treatment with PEI1200-CDots under the same conditions only resulted in 1/82 log reduction in viable cell number. The much higher antibacterial efficiency of PEI600-CDots could be rationalized such that the corresponding thinner passivation layer might allow the ROS to act more effectively on the bacterial cells for the observed more effective antibacterial function .
For gene targeting, homology arms to human Bub1 (BUB1) gene were amplified from RPE1 cell genomic DNA. Targeting constructs allowing the insertion of an EGFP tag C-terminal to Bub1 were assembled by 4-piece ligation in a NotI-digested pAAV vector. Recombinant adenovirus-associated virus (rAAV) particles were generated as previously described (Berdougo et al, 2009). RPE1 cells were infected with 3 ml of viral supernatant for 48 hr and then expanded into fresh medium for an additional 48 hr. FACS sorting was used to select EGFP-positive cells, as previously described (Collin et al, 2021). To facilitate detection of fluorescence at mitotic stages, cells were synchronized with RO-3306 (10 μM) for 18 hr and released into nocodazole (50 nM) for 2 hr, before they were trypsinized and subjected to sorting in the continued presence of nocodazole (10 nM). Infected or uninfected cells were filtered (30 μm, Partec) and EGFP-positive cells (488 excitation, 514/30 emission filter) were isolated on an Aria IIIu (BD) cell sorter by selecting the 514/30 channel against a 585/42 filter detecting cellular autofluorescence. Single cells were sorted into 96-well plates filled with conditioned medium and positive clones screened for by fluorescence microscopy.
Faster loom speeds were always accompanied by larger final dot sizes (see Materials and Methods). The increased response to faster loom speeds was not due to the larger final dot sizes, however.

To perform fiber photometry recordings from VTA DA neurons, we expressed the calcium indicator GCaMP6f in DA neurons by crossing Dat-Cre mice with Ai148D mice (see Materials and methods; Engelhard et al, 2021). We targeted the recording fiber to either medial or lateral VTA (Figure 3A–C, Figure 3—figure supplement 1A; medial VTA group: n = 10 mice, lateral VTA group: n = 11 mice). We chose these subregions because of the electrophysiological, anatomical and functional evidence that there is a medial/lateral distinction within the VTA (Lammel et al, 2021; Beier et al, 2021; Yang et al, 2021; Engelhard et al, 2021; de Jong et al, 2021). We found that both regions showed increased activity to reward (Figure 3—figure supplement 2).

Again, primary blasts from different patients displayed a high degree of heterogeneity of drug response. The median CI values obtained for lestaurtinib/AraC, PKC412/AraC,and lestaurtinib/PKC412 were 0/73, 0/78 and 0/96, respectively, suggesting an overall minor degree of synergy when the FLT3 inhibitors were combined with AraC and an approximately additive effect when the 2 FLT3 inhibitors were used in tandem. To determine statistical significance, the distribution of the study population relative to a CI cutoff value of 1 was compared with an equally sized hypothetical “additive” population with a median CI of 1 using the V2 test.

Indeed, the connecting bridge transmitting the force developed by the cytoskeleton to actively reshape the PM and pull it inward can only support a finite amount of force, above which it breaks and actin polymerization becomes futile.
Using the CD-ROM With Automounters On many newer systems, the CD-ROM is automatically mounted when you insert it. In this case, you simply need to run the installation program. NOTE: If you intend to do backups to CD, DVD or REV media, we highly recommend that you disable any automount daemons on your system. They will try to mount your backup media when inserted, with unpredictable results. Newer Linux systems have both automount and autorun capabilities when running under GUI desktops. If you are logged in as root under the KDE desktop and insert the CD-ROM, the BackupEDGE installation menu will appear automatically if autorun is enabled (or you will be prompted to confirm that you want to autorun the install program).
CMR is a psychophysical phenomenon in which the detection of an on-frequency signal (“signal”) in the presence of modulated on-frequency masker (“masker”) is improved by adding off-frequency sounds (“sidebands”) that are comodulated with the masker (Hall et al, 1984). T-Stellate cells have been shown to exhibit CMR in their spike trains, and this sensitivity can be explained by a simple circuit consisting of a wideband inhibitory input (such as from D-stellate cells) that converges onto cells receiving narrowband excitation from the AN (Pressnitzer et al, 2001).
Endocytosis is the process by which eukaryotic cells internalize material and information from their environment and recycle plasma membrane (PM) lipids, trafficking proteins, and cell-surface receptors. Membrane remodeling by a well-established sequence of protein complexes (Fig. S1 A) is essential to form endocytic buds that will internalize material (Kaksonen and Roux, 2021). Thus, the PM can be considered as a core part of the endocytic machinery. It is now broadly accepted that physical forces, in particular PM tension, participate in the regulation of the balance between exocytosis and endocytosis in various systems (Dai and Sheetz, 1995; Gauthier et al, 2021). Functioning in a homeostatic feedback loop, the opposing effects of endocytosis and exocytosis on PM area is generally believed to enable cells to keep tension close to a set point (Morris and Homann, 2001; Apodaca, 2002; Fernandez-Sanchez et al, 2021). Additionally, PM tension was shown to regulate specific steps of the endocytosis process, including clathrin pit formation by varying the membrane budding energy (Boulant et al, 2021; Saleem et al, 2021) and membrane fission by dynamin (Morlot et al, 2021). These tensile forces, depending on the geometry of the bud, constitute either a basal constraint that the cell machinery has to counteract or a driving force in order to reshape the PM and form the endocytic vesicle.
